摘 要:目的:揭示补脑止痫散对戊四氮(PTZ)慢性点燃癫痫大鼠模型c-fos、c-jun蛋白表达的影响。方法:SD大鼠60只,随机分为正常对照组、模型组、丙戊酸钠组、补脑止痫散小剂量组、补脑止痫散中剂量组、补脑止痫散高剂量组。采用PTZ小剂量连续腹腔注射方法,建立慢性点燃癫痫大鼠模型,获得Ⅲ级以上发作后,立即停止给药。采用免疫组化法检测各组大鼠海马区c-fos、c-jun蛋白表达。结果:除对照组无痫样发作外,其余各组全部出现Racine分级Ⅲ-Ⅴ痫样发作。通过免疫组化检测海马区的c-fos、c-jun蛋白表达,丙戊酸钠组、补脑止痫散高剂量组c-fos、c-jun蛋白表达明显下调。二者与模型组比较,差异有统计学意义(P<0.05)。结论:补脑止痫散能对戊四氮慢性点燃癫痫大鼠模型海马区内即刻早期基因有明显的调控作用,从而治疗癫痫、保护脑组织,减轻由癫痫导致的损伤。Objective:To study the expressive effects on c -los and c -jun protein in the brain of Pentylenetet- razole (PTZ) -kindled chronic epileptic rats. Methods: 60 healthy SD rats were randomly divided into six groups, and they were control group(10) , epileptic rat model group (10), Sodium valproate group (10) , low dose of Bunao Zhixian powder therapy group( 10), middle dose of Bunao Zhixian powder therapy group(10) and high dose of Bunao Zhixian powder therapy group (10). Rat kindling model of epilepsy was established by PTZ - injection. By immuno histochemistry methodwe detected the protein expression of c - fos c - jun in hippocampus of rats. Results:The occurrence and seizure level were evaluated by using Racine's method. The rats with 111 to V seizure level were chosen for the following studies. The expression of c - fos and c - jun pro- tein decreased significantly between the Sodium valproate group and the high dose of Bunao Zhixian powder therapy group. There were significant differences by comparing with model group (P 〈 0.05 ). Conclusion : Bu- nao Zhixian powder plays a significantly regulative role in immediate - early gene, in order to treat epilepsy, to protect the brain tissue and reduce the damage caused by epilepsy.
