摘  要:目的:采用单味药洋金花细粉对PD模型大鼠进行治疗,并探讨其作用机制,从而为中药治疗帕金森病提供一条新的思路。方法:采用脑立体定位注射6-OHDA建立大鼠PD模型,阿朴吗啡(Apo-morphine)诱导进行旋转行为观察,将模型成功大鼠随机分为洋金花高、中、低剂量组、美多芭组及模型对照组,同时设假手术组作为对照。观察其对超氧化物歧化酶(SOD)、丙二醛(MDA)、谷胱甘肽(GSH)、谷胱甘肽过氧化物酶(GSH-Px)变化的影响。结果:给药3周后,各治疗组纹状体内MDA含量均有明显降低(P<0.01),以美多芭组、洋金花高、中剂量组降低显著,三组之间比较无显著性差别(P>0.05);各治疗组纹状体内SOD含量均有明显增加(P<0.05),以美多芭组、洋金花高剂量组增加显著,两组之间比较无显著性差别(P>0.05);各治疗组纹状体内GSH、GSH-Px均有明显增加(P<0.05),其中以洋金花中剂量组增加最明显。结论:洋金花可以降低PD模型大鼠纹状体MDA含量,提高GSH、GSH-Px、SOD的含量,可能是通过减轻脂质过氧化反应,增强抗氧化防御机制,减少氧化应激,进而保护神经元,起到治疗PD的作用。Objective : Use single herbs Yangjinhua fine powder to treat rat model with PD to explore some of its mechanism, so as to provide a new way of Chinese medicine to treat Parkinson's disease. Method: Use ster- eotaxic injection of 6 - OHDA to establish rat models with PD, apomorphine induced rotational behavior obser- vation, the model rats were randomly divided into Yangjinhua high , medium and low -dose groups, the Madopar group and the model control group, at the same time set up the sham group as comparison. Observe the impact of changes of superoxide dismutase ( SOD), malonic aldehyde (MDA) and glutathione ( GSH), glutathione peroxidase (GSH -Px). Results: After administration 3W, the striatum of MDA content of each treatment group was significantly lower ( P 〈 0.01 ), to the Madopar group, the Yangjinhua high and medium dose groups decreased significantly, among the three groups there was no significant difference (P 〉 0.05 ) ; the SOD content of each treatment group Striatum was significantly increased ( P 〈 0. 05 ). to the Madonar group, the Yangjinhua high dose group increased significantly, between the two groups there was no significant difference ( P 〉 O. 05 ) ; Each treatment group striatum, GSH, GSH - Px were significantly increased ( P〈 0. 05 ), with the most obvious increase in the Yangjinhua medium dose group. Conclusion : Yangjinhua can re- duce the content of PD model rat striatum MDA, improve the content of GSH, GSH - Px, SOD, the mecha- nism may be by reducing lipid peroxidation, enhancing the antioxidant defense mechanism, reducing the oxi- dative stress, thereby to protect the neurons, and plays a role in the treatment of PD.
