摘  要:目的:研究氯喹对鼠伤寒沙门菌诱导巨噬细胞(macrophage,MΦ)自噬的影响。方法:确定氯喹干预感染模型的工作浓度,应用该浓度下的氯喹分别干预携带毒力质粒的野生株χ3306和消除了毒力质粒的突变株χ3337与小鼠腹腔MΦJ774A.1的感染模型,探讨氯喹影响细菌与MΦJ774A.1自噬的机制。结果:20μmol/L氯喹单独作用6 h对细胞和细菌无显著影响;但干预感染模型后,却能阻断自噬。结论:鼠伤寒沙门菌毒力质粒参与了自噬早期过程。Objective: To investigate the effect of chloroquine on the autophagy of macrophage induced by Salmonella enterica serovar Typhimnrium (STM). Methods: First of all, we found out the appropriate concentration of chloroquine, and then we used a strain of STM infecting mouse peritoneal macrophage(J774A.1) model to discuss the mechanism by chloroquine intervention. In the model, X3306 (a STM strain containing the virulent plasmid) and X3337(a STM strain without the virulent plasmid) were used. Results: After six hours' interaction with the 20 υmol/L chloroquine, the effect on the host cells and the bacteria was not noticeable, but after the intervention of chloroquine, the autophagy of the host cells was blocked. Conclus/on: The virulent plasmid which carried by STM influences the early stage of autophagy.
