摘  要:目的:建立大鼠单侧红核脊髓束损伤模型并利用体视学方法评估红核中枢性损伤情况。方法:成年SD大鼠30只,随机分为正常组、红核脊髓束(rubrospinal tract,RST)损伤组和假手术组。RST损伤组选择性切断SD大鼠脊髓C。左侧背外侧索制作单侧RST横断损伤模型,术后1、2、8周对各组大鼠采用前肢支撑探测实验进行行为学评价.用Fluoro—mby(FR)逆行荧光示踪及体视学定量分析法观察红核神经元的形态及数目变化。结果:前肢支撑探测实验结果显示各时间点RST损伤组大鼠左前肢使用率均低于正常组和假手术组(P〈0.05);FR染色结果可见RST损伤组大鼠中脑右侧FR阳性红核神经元数目较左侧明显减少,细胞荧光较弱,细胞突起较少,且短细。与其余两组比较RST损伤组大鼠右侧中脑FR阳性红核神经元计数也明显减少fP〈0.05)。结论:选择性切断SD大鼠脊髓背外侧索能成功制备大鼠单侧RST损伤模型.应用体视学方法可以精确评估传导束损伤引发的继发性红核神经元改变.Objective: To establish an animal model of rubrospinal tract(RST) injury and estimate it accurately by sterology for central nervous sysytem regeneration research on rat. Methods: 30 adult rats were selected in this experiment and were randomly dividied into three groups, namely the normal group, sham operatin group and RST injury group. Unilateral RST injury model was established by transecting the left dorsolateral funiculus at C3-4 segment of rat spinal cord on RST injury group. The behavior was evaluated with forelimb probing experiment after 1 w, 2 w and 8 w. Red nucleus neurons were shown by fluoro-ruby(FR) and analyzed by using disector. Results: The forelimb probing test showed a lower use rate of left forelimb in the RST injury group rats than that of other groups with statistical significance(P〈0.05). There had more remarkable FR positive neurons and their projections in left red nucleus than that in the right one in RST injury group. There also had a significant reduction of FR positive neurons in right red nucleus in RST injury group compared to the normal group and the sham operation group(P〈0.05). Conclusion: The RST injury model could be established by selectively cutting off the dorsolateral funiculus, and the change of the red nucleus neurons could be evaluated by sterology accurately.
