摘  要:海水养殖生物病毒病是世界性难题,淋巴囊肿病毒中国分离株(LCDV-cn)是牙鲆(Paralichthys olivaceus)淋巴囊肿病的病原。以mcp基因为靶基因,设计siRNA-mcp1、siRNA-mcp2、siRNA-mcp3等3条siRNA序列,研究RNA干扰(RNAi)对淋巴囊肿病毒在牙鲆体内复制的影响。结果表明,牙鲆体内注入siRNA-mcp1和siRNA-mcp3后,mcp基因的表达有所下调;统计分析表明,注入siRNA-mcp1后,mcp基因的表达下调明显,具有显著性差异(p<0.05),注入siRNA-mcp2、siRNA-mcp3后,mcp基因的表达下调不明显,差异不显著(p>0.05)。本实验中初步实现了RNAi对LCDV-cn复制的抑制作用。Lymphocystis disease virus- Chinese isolate (LCDV-cn) is the pathogen of lymphocystis disease in Paralichthys olivaceus, causing a huge loss in flounder aquaculture. Controlling viral infection in aquiculture is a technological challenge worldwide, and RNAi could be a way of preventing viral infection of cultured animals. In this study, the mcp gene as a target gene, three siRNA sequences (siRNA-mcpl, siRNA-mcp2 and siRNA-.mcp3) were designed and used to study the effect of RNA interference (RNAi) on the replication of LCDV-cn in the body of Paralichthys olivaceus. The experimental results showed that the expression of the mcp gene had the down-regulation after siRNA-mcpl and siRNA-mcp3 had been injected in the body of Paralichthys olivaceus. Statistical analysis showed that the down expression of the mcp gene was significantly different (p 〈 0.05) after the injection of siRNA-mcpl in the body of Paralichthys olivaceus, but did not occur (p 〉 0.05) after the injection of siRNA-mcp2 and siRNA-mcp3. The preliminary realization of the inhibition of RNAi on the replication of LCDV-cn had been acquired in this study.
