摘 要:目的研究豹皮樟总黄酮(TFLC)对急性炎症的预防作用并探讨其是否与内质网应激(ERS)调控的抗炎作用有关。方法采用角叉菜胶皮下注射,诱导大鼠足肿胀模型,观察TFLC的抗炎作用,病理学分析肿胀足爪部位变化,并检测大鼠血清和足爪组织中一氧化氮(NO)、丙二醛(MDA)、前列腺素E2(PGE2)、肿瘤坏死因子-α(TNF-α)和白介素-10(IL-10)的含量。Western blot方法分析ERS相关蛋白CCAAT增强子结合蛋白同源蛋白(CHOP)和葡萄糖调节蛋白78(GRP 78)的变化。结果 TFLC可以明显抑制角叉菜胶所致的大鼠足肿胀,并且可以降低血清以及炎症组织渗出液中NO、MDA、PGE2、TNF-α的含量,同时升高IL-10的表达。Western blot结果显示,TFLC可以明显降低腹腔巨噬细胞中CHOP和GRP 78的表达。结论 TFLC对角叉菜胶诱导的大鼠足肿胀有明显的抑制作用,其作用机制可能通过影响炎症因子分泌及抑制ERS诱导的炎症损伤。Objective To obseve the prophylactic effect of total flavonoids from Litsea coreana Levl. (TFLC) in carrageenan-indueed paw edema in rats and explore Endoplasmic Reticulum Stress (ERS)-mediated anti-inflammation of TFLC. Methods Thirty rats were given TFLC (50, 100, 200 mg/kg) for 5 days and then the rats received a subplantar injection of carrageenan in the left hind paw. After the treatment, inflammation was analyzed by paw swelling and histopathology. Malondialdehyde ( MDA), nitric oxide ( NO), prostaglandin E2 ( PGE2 ), tumor necrosis factor α (TNF-α)and interleukin 10 (IL-10) in serum and paw tissue were studied. Western blot was used to analyze the expression of CCAAT/enhaneer binding protein homologous protein (CHOP) and Glucose regulated protein 78 ( GRP 78 ). Results Our results showed that TFLC had good effects on carrageenan-induced paw edema. Additionally,the expression of MDA, NO, PGE2 and TNF-α were higher in model rats and then decreased after given TFLC, on the contrary to IL-10. Western blot analysis showed that TFLC could reduce expression of CHOP and GRP 78. Conclusion These results reveal that TFLC could inhibit carrageenan-induced paw edema and the mechanisms might be related to regulate the secretion of inflammatory cytokines and suppress ER stress.
