摘 要:目的比较不同培养方法对冻融人卵巢组织内卵泡生长发育的影响。方法实验分3组:冻融的人卵巢组织单独培养组(A组);冻融的人卵巢组织与人早孕蜕膜单层细胞共培养组(B组);Matrigel培养基培养的冻融的人卵巢组织组(C组)。冻融人卵巢组织在不同培养条件下培养14 d后,固定并检测各组卵巢组织中总体卵泡存活率(TFSR)及卵泡生长率(GFR)。结果与B、C组比较,A组TFSR和GFR均降低,差异有统计学意义(P<0.05)。与C组比较,B组TFSR和GFR无明显变化,差异无统计学意义。结论冻融卵巢组织中的原始卵泡在不同体外培养体系中有不同程度的生长;冻融卵巢组织与人早孕蜕膜细胞共培养或Matri-gel培养基培养相对于单独培养更有利于卵泡存活和生长;但这两种培养方法之间没有明显差异。Objective To compare the influences on the development of the follicles by different culture system for thawed human ovary tissues. Methods Three groups in the experiment:the thawed ovarian tissues were cultured alone( group A) ;coculturing the thawed ovarian tissues with the decidua monolayer cells (group B ) ;culturing the thawed ovarian tissues with Matrigel-coated flask( group C ) ;after 14 days of culture ,the total follicle survival rate (TFSR) and the grow follicle rate (GFR) among the groups were compared. Results There were statistical differences on TFSR(P 〈0.05 ) and GFR (P 〈 0. 05)between group A and B; compared with group A, group C had the same results. Comparing the two indices between B and C, the differences were not significant. Conclusion The primordial follicles developed in different speed in different culture systems. Co-cuhure of the thawed ovarian tissues with the decidua monolayer cells or Matrigel are better than single method. But there are no obvious advanta- ges and disadvantages between the two culture system.
