摘 要:以细毡毛忍冬叶片为外植体建立了离体再生体系,在此基础上,以叶片诱导出愈伤组织,用秋水仙素诱导其同源四倍体。结果表明,秋水仙素浓度为0.2g·L-1,液体培养48h,四倍体诱导率最高为15.0%。四倍体细毡毛忍冬生长缓慢,植株节间短,茎干粗壮,叶片颜色较二倍体浓绿,叶形指数减少,保卫细胞增大,气孔密度减少,绿原酸含量增加。To establish an effective leaf generation system and induce stable tetraploid ofLonicera similis, calluses were induced by using leaves ofL. similis as explants and calluses were employed to establish the technical system of autotetraploid induction in L. similis by colchicines. The results showed that the highest induction rate of tetraploid, 15.0%, was obtained from the callus cultured in the liquid medium supplemented with 0.2 g.L-1 colchicine for 48 h. The plants grew slowly and showed the general characteristics of tetraploids, such as shorter internodal, thicker stem, deeper leaf, smaller leaf index, larger guard cells and less stomata density. The contents of chlorogenic acid of tetraploid plants were higher than diploid plants.
