摘  要:目的研究基质相互作用分子1(stromal interaction molecule 1,STIM1)和钙释放激活钙通道调节分子1(calcium release-activated calcium channel modulator 1,ORAI1/CRACM1)在外伤性癫痫大鼠脑组织皮层中的表达变化,探讨其与外伤性癫痫的关系及其意义。方法84只成年SD大鼠随机分为对照组(42只)和实验组(42只),制作FeCl3外伤性癫痫模型,实验组予皮层注射100mmol/L FeCl35μL,对照组皮层注射等量生理盐水,观察记录大鼠伤后癫痫发作的行为学表现,伤后不同时间点(6h、24h、72h、7d、14d、21d、28d)随机取6只大鼠伤侧皮层脑组织,用荧光定量PCR、Western blot和免疫组化技术检测STIM1和ORAI1的mRNA和蛋白的表达。结果实验组伤后出现典型的癫痫发作,制模成功率达84.0%,对照组未见癫痫发作;实验组STIM1、ORAI1的mRNA表达均较对照组显著增高(P<0.05),72h为表达高峰(P<0.05);实验组STIM1、ORAI1在伤后6h的蛋白表达与对照组比较无统计学差异(P>0.05),此后6个时间点的表达与对照组比较均显著增高(P<0.05),72h为表达高峰(P<0.05)。结论STIM1、ORAI1在外伤性癫痫大鼠皮层脑组织中表达显著上调,钙释放激活的钙(calcium re lease-activated calcium,CRAC)通道可能与外伤后癫痫的形成有关。Objective To investigate the expression of stromal interaction molecule (STIM1) and calcium release-activated calcium channel modulatorl (ORAI1/CRACM1) in cortical brain tissue of post-traumatic epilepsy rats, and to explore the possible role in post-traumatic epilepsy. Methods Eighty four adult SD rats were randomly divided into two groups: control group (42) and experimental group (42). The rats in experimental group received an intracortical injection of lO0mmol/L FeC13 51M wheras the animals in the control group received an intracortical injection with equal saline. Rat models of epilepsy were established by intracortical microinjection of ferric chloride. The occurrence of seizures was observed and recorded continuously 6 h later. Six rats were randomly selected at each time points ( 6 h, 24 h, 72 h, 7 d, 14 d, 21 d and 28 d) after epileptic seizure. The mRNA and protein expression levels of STIM1 and ORAI1 were detected by using Real-time PCR, Western blot and immunohistochemistry, respectively. Results Typical seizures were observed in experimental group, and the attack rate was 84.0 %. Protein and mRNA expression levels of STIM1 and ORAI1 in the experimental group were increased and peaked at 72 h following injection of FeCh Expression levels of STIMI and ORAI1 mRNA in the experimental group were significantly higher than those in the control at each time points (P 〈0.05), and peaked at 72h (P 〈0.05).There was no statistical difference in the expression levels of STIM1 and ORAI1 protein between experimental and the control groups at 6 h (P 〉0.05);Expression levels of STIM 1 and ORAI1 protein in experimental group were significantly higher than those in the control at the latter six time points (P 〈0.05),and peaked at 72h (P 〈0.05). Conclusions Expression levels of STIM1 and ORAI1 are increased in the cortex of post-traumatic epilepsy rats, indicating that CRAC may contribute to the formation of post-traumatic epilepsy.
