SPIO标记对成骨样分化细胞IRP1基因及蛋白表达的影响

摘　　要：【目的】探讨超顺磁性氧化铁(SPIO)标记大鼠脂肪间充质干细胞(RADSC)后,其成骨样分化细胞中铁调节蛋白1(IRP1)基因及蛋白表达的变化。【方法】本实验采用转染剂多聚赖氨酸(PLL)介导SPIO标记RADSC,然后将其诱导为成骨样分化细胞,分别在诱导后0、7、14、21 d进行RT-PCR及Western blot检测其IRP1基因及蛋白的表达。【结果】成骨样分化细胞IRP1 mRNA表达水平在SPIO标记与未标记组之间差异均无统计学显著性意义(F=1.571,P=0.239);IRP1蛋白表达水平在标记与未标记组之间差异均无统计学显著性意义(F=0.547,P=0.500)。【结论】SPIO标记并未对成骨样分化细胞的IRP1基因及IRP1蛋白表达产生明显影响。[Objective]To evaluate the effects of SPIO labeling on the IRP1 gene and protein expression in osteoblast-like differentiated cells. [Methods] To label the RADSC with SPIO and transfection agent PLL, then induce SPIO labeled and unlabeled RADSC into osteoblastic differentiation cells. Real-time quantitative PCR and Western blot was used to exam the expression level of IRP1 gene and protein on the 0 day, 7 day, 14 day, and 21day after induction. [Result] The expression of IRP1 gene of osteoblast- like cells had not changed significant after labeling （F = 1.571, P = 0.239）. The expression of IRP1 protein of osteoblast-like ceils had not changed significant after labeling （F = 0.547, P = 0.500）. [Conclusion] Osteoblast-Like cells were not effected by SPIO not only in the aspect of IRP1 gene, but also in IRP1 protein.
