摘  要:目的:观察大黄素(EMO)对HepG2肿瘤细胞的形态、增殖以及v-Src蛋白表达的影响。方法:采用光学显微镜观察大黄素处理后肝癌细胞HepG2的形态变化,MTT法测定细胞的增殖作用,酶联免疫吸附(ELISA)法检测HepG2细胞培养上清中v-Src蛋白的表达水平。结果:大黄素后能明显抑制HepG2细胞的增殖作用,IC50值为43.77ug/mL。经不同浓度大黄素处理的HepG2细胞中v-Src蛋白的表达显著降低,呈现剂量相关性。结论:大黄素能抑制HepG2细胞的增殖,降低v-Src蛋白表达。Objective:The present study assessed the effect emodin(EMO) on HepG2 cell line morphology growth and V-Src protein ex- pression. Methods:To observe the change of the cells morphology structure by light microscope. To clarify the inhibitory effect of EMO on HepG2 cell line, MIT assay was performed. While ELISA assay was used to determine the V-Src protein expression level in HepG2 cell line culture superuatant.Results:HepG2 cell line showed dose-dependence when significantly reduced by EMO. IC~ was 43.77ug/ ml. The high-level expressions of V-Src protein in HepG2 cell line dealing with different concentrations of EMO were significantly dose-related. Conclusions:Our results demonstrated that EMO can inhibit the V-Sre protein expressions in HepG2 cell line and promote apoptosis of cancer cells.
