坤宁安对围绝经期模型大鼠性腺形态影响

摘　　要：目的：观察坤宁安对围绝经期模型大鼠性腺形态的影响,探讨坤宁安治疗围绝经期综合征的疗效及作用机制。方法：13~16月龄的Wistar大鼠40只,以自然老化法的方法建立围绝经大鼠模型,随机分为坤宁安高（n=10）、低剂量组（n=10）、西药对照组（n=10）与模型组（n=10）;另选3~5月龄的Wistar大鼠10只作为青年对照组。坤宁安高、低剂量组分别予以坤宁安3.75、0.94 g/kg日1次灌胃治疗,西药对照组予以倍美力0.065 mg/kg日1次灌胃治疗,模型组和青年对照组均予以等体积的生理盐水灌胃,连续4周。治疗结束后称取各组卵巢及子宫重量,苏木素-伊红（HE）染色法观察各组卵巢形态变化。结果：与青年对照组相比是显著降低的（P〈0.05）。卵巢HE染色显示与模型组相比,坤宁安高、低剂量组卵巢闭锁卵泡显著减少,黄体数量增多,其中高剂量组疗效优于低剂量组。结论：坤宁安可显著增加围绝经期综合征模型大鼠卵巢及子宫重量,减少闭锁卵泡的数量,增加黄体数量,改善卵巢功能,可用来防治围绝经综合征。Objective ： To observe the effect of traditional Chinese medicine Kunning＇an （ KNA ） on genitical gland function and the weight of the womb and the ovary of perimenopausal rats. Methods ： 40 natural senile female Wistar rats were randomly divided into model group, high dose group of KNA, low dose group of KNA, western medicine control group. And another 10 young female rats were as compared group. The changes of genitical gland function and the weights of the womb and the ovary among the 5 groups were compared. Results ： Compared with model group, the weights of the womb and the ovary of young control group were much higher （ P〈0.01 ）, the weights of the womb and the ovary of high and low dose groups of KNA and western medicine control group were higher （ P〈0.05 ）, but it was lower than those in young control group （P〈0.05）. The result was statistically significant. KNA can also improve the pathological change of ovary. Conclusions ： KNA Pill can increase the weights of the womb and the ovary obviously and reduce the number of atretic follicles and improve the pathological change of overy effectively of perimenopausal rats. Consequently, it can be used for perimenopausal syndrome.
