摘 要:目的探讨米诺环素对大鼠肢体缺血-再灌注继发肺损伤的影响。方法 SD成年雄性大鼠30只,随机分为三组(n=10),采用乙醚麻醉后,N组和M组大鼠经尾静脉注入生理盐水1.0ml,C组注入米诺环素10mg/kg。M组和C组分别结扎大鼠双下肢3h,再灌注4h制作缺血-再灌注模型。切除右肺,上叶测量湿干比,中叶测定IL-6含量;左肺下叶免疫组化S-P法观察基质金属蛋白酶(MMP)-2和MMP-9在肺组织的着色及灰度对比。结果右肺上叶湿干比、左肺下叶MMP-2和MMP-9表达以及匀浆IL-6含量均为M组>C组>N组(P<0.01)。结论米诺环素可通过抑制炎性反应减轻大鼠肢体缺血-再灌注所造成的肺损伤。Objective To explore the effect of minocyeline in lung tissue after limbs ischemia- reperfusion in rats. Methods Thirty healthy SD rats were randomly divided into 3 groups (n= 10) : group M,group N and group C. Group N and group M received iv. normal sodium 1.0 ml. Group C received iv. minocycline 10 rng/kg (about 1.0 ml). Group M and group C received lower limb ligation for 3 h and reperfusion for 4 h. Right lung wet-dry ratio, levels of MMP-2, MMP-9 and IL-6 were investigated in the left lung. Results The w/d ratio, the expressions of MMP-2, MMP-9, and the level of tissue IL-6 was increased by limb ischemia-repeffusion with the sequence group M ~ C ~ N (P ~ 0.01 ). Conclusion Minocycline can protect the lung against ischemia-repeffusion injury by inhibiting inflammatory response.
