摘 要:目的探讨高压氧对脊髓损伤后Bcl-2蛋白表达的影响及意义。方法将54只健康成年雌性Wister大鼠随机分为假手术组6只、损伤组和实验组各24只。采用Allen′s法制作大鼠脊髓损伤模型,术后实验组进行高压氧治疗,损伤组和假手术组动物常规饲养。分别在各个时间点取材,进行常规HE和免疫组织化学染色,观察脊髓组织学及Bcl-2蛋白表达情况。结果HE染色可见损伤组脊髓组织结构紊乱,组织出血、水肿、空泡变性。实验组脊髓结构相对完整,组织水肿程度减轻。免疫组化染色可见假手术组偶见Bcl-2阳性表达,损伤组和实验组均可见Bcl-2表达,实验组Bcl-2阳性表达较高,与损伤组相比具有显著性差异(P<0.05)。结论高压氧可能通过增加脊髓损伤后Bcl-2蛋白表达抑制神经细胞凋亡,从而对脊髓损伤起到治疗作用。Objective To investigate the effect and significance of hyperbaric oxygen (HBO) on expression of Bcl-2 protein after spinal cord injury in rats. Methods Fifty-four female adult Wistar rats, weighing between 250 and 300 g, were randomly divided into sham opera- tion group ,spinal cord injured group and experimental group. The spinal cord injury was induced by Allen's method. Rats in experimental group were treated with HBO, while the rats in other two groups were rearing under conventional condition. Animals were sacrificed at differ- ent time points and tissues were collected. The expression of Bcl-2 protein and the change of spinal cord histology were detected by immuno- chemistry and hematoxylin-eosin (HE) staining. Results The HE staining results of histology showed structural disorder of spinal cord, hemorrhage,edema,and a number of vacuolation in injured spinal cord;the above mentioned symptoms were completely relieved in HBO group but not in sham operation group. The immunochemistry results showed only a few cells in sham operation group were positive for Bcl- 2, which was widely visible in injured group and experimental group;significantly increased number of Bcl-2-positive cells were observed in experimental group compared with the spinal cord injured group (P 〈 0.05). Conclusion The HBO inhibits the apoptosis of neuron by in- creasing the expression of Bcl-2 protein, and thus play a therapeutic role for spinal cord injury.
