摘 要:目的评价羟基磷灰石(HA)涂层的两种合金Mg-4.0Zn-0.5Sr(Wt.%)和Mg-4.0Zn-1.5Sr(Wt.%)的细胞毒性和生物相容性。方法先制备HA涂层Mg-4.0Zn-0.5Sr和Mg-4.0Zn-1.5Sr两种合金,再进行浸提液的制备、测定其镁离子浓度、培养小鼠成纤维细胞L929、测定MTT细胞活力及溶血率。结果 Mg-4.0Zn-1.5Sr合金浸提液的pH值高于Mg-4.0Zn-0.5Sr合金(P<0.05),但镁离子浓度低于Mg-4.0Zn-0.5Sr合金(P<0.05),相对增殖度无统计学差异(P>0.05),但溶血率低于Mg-4.0Zn-0.5Sr合金(P<0.05)。24、48、72 h培养后,对L929细胞的计数显示,两合金组细胞数量减低,但与对照组比较无统计学差异(P>0.05)。结论 Mg-4.0Zn-0.5Sr和Mg-4.0Zn-1.5Sr两种镁合金无细胞毒性,生物相容性较好,有进一步研究应用的价值。Objective To appraise cytotoxicity and biocompatibility of Mg-4.0Zn-0.5Sr alloy and Mg-4.0Zn-l.5Sr alloy,respectively. Methods Both of the magnesium alloy with hydroxyapatite (HA) coated were prepared firstly,then the extracts of the alloy samples were picked-up to determine the magnesium ion concentration, and through the cultivated of the fibroblasts L929, the cell vigor of MTF colorimetry assessment and hemolysis ratio were obtained. Results The pH of the extract of Mg-g.0Zn-l.5Sr alloy is higher than that of Mg-4.0Zn- 0.5Sr alloy (P 〈 0.05 ), but the magnesium ion concentration of Mg-4.0Zn-l.5Sr alloy is lower than that of Mg-4.0Zn-0.5Sr alloy, as well as both of the relative growth rate (RGR) are similar and the hemolysis ratio of Mg-4.0Zn-l.5Sr alloy is lower than that of Mg-4.0Zn-0.5Sr al- loy (P 〈 0.05 ). Through 24 h,48 h and 72 h cultivated,the count of L929 cell is shown that both counts of the alloys are decreased slightly and there are not marked difference compared with the comparison group. Conclusion Because the cytotoxicity of both Mg-4.0Zn-0.5Sr al- loy and Mg-4.0Zn-l.5Sr alloy is not shown and there is a better biocompatibility,the further study is needed.
