凝结芽孢杆菌产芽孢发酵条件的优化

摘　　要：通过单因素试验和正交试验,对凝结芽孢杆菌NJ-2的产芽孢发酵条件进行了优化。优化后的培养基组成为:玉米面30 g/L,豆粕27 g/L,(NH4)2SO413 g/L,麸皮30 g/L,MgSO41 g/L,Na2HPO410 g/L,NaH2PO40.5 g/L,pH 7.0;最优的培养条件为:温度40℃,初始pH 7.0,转速180 r/min,装液量100 mL(500 mL的三角瓶),接种量6%(V/V),发酵时间36 h。在该优化条件下,芽孢形成率达到85%。The single factor and orthogonal tests were carried out to optimize the spore-forming conditions ot kloctUus coagulan NJ-2.The optimal medium were as follows：corn meal 30 g/L,soybean 27 g/L, （NH4）2SO4 13 g/L,bran 30 g/L, MgSO4 1 g/L,Na2HPO4 10 g/L, NaH2PO4 0.5 g/L,The optimal culture conditions were as follows ：temperature 40 ℃ ,initial pH 7.0, rotation speed 180 r/mim, volume of shaking flask 100 mL/500 mL,the inoculum was 6% （V/V）and the fementa- tion time 36 h.Under above conditions, the sporulation rate reached 85%.
