人血清白蛋白基因乳腺表达载体在奶山羊乳腺中的瞬间表达

摘　　要：构建了包括人血清白蛋白 ( h ALB)基因的 c DNA全序列、内含子 1以及山羊β-酪蛋白基因启动子和 5′端上游调控序列在内的乳腺表达载体 pc DNA3.1 -GCALBm,采用直接注射法将其导入哺乳期的奶山羊乳腺组织 ,然后通过放射免疫法测定乳汁中 h ALB的含量。结果显示 ,5头受试奶山羊的乳汁中有明显的 h ALB表达 ,注射后第 2天放免测定值从注射前的 4 .0 mg/L左右分别上升至 62 .2、59.2、36.2、4 2 .3、1 7.5mg/L,并持续表达在 2周以上 ,其中 1头受试羊乳汁中 h ALB的表达量维持在 80 mg/L左右 ,并超过了 1 0 0 d。而在乳腺组织中注射 pc DNA3.1空质粒或生理盐水的 4头对照奶山羊的乳汁中 ,放免测定值无变化。A mammary gland expression vector pcDNA 3 1 GCALBm containing the full sequence of human serum albumin(hALB) cDNA,intron 1 as well as goat β casein gene promoter with 5′ upstream regulatory sequence was constructed.The vector was directly injected into the goat mammary gland during the lactating period.The milk was collected for determination of hALB content with radioimmunoassay.The results showed that the hALB content in the milk of 5 tested goats were significantly increased.The quantity from 4.0 mg/L before injection rose up to 62.2,59.2,36.2,42.3 and 17.5 mg/L,respectively,on the 2nd day after injection.This expression level persisted for two weeks.The hALB content in the milk of one tested goat reached to about 80 mg/L for over 100 days,while the value of radioimmunoassay of the control goats injected with null pcDNA 3.1 vector or saline solution showed no any changes.The results in this paper provide valuable information for creating hALB transgenic bioreactors.
