摘 要:40日龄艾维茵肉鸡 30只 ,随机分为 3组 ,分别进行了静注、肌注及内服沙拉沙星 ( 1 0 mg/kg)的药动学研究。血浆样品经甲醇沉淀血浆蛋白 ,高速离心 ,用反相高效液相色谱法测定鸡血浆中沙拉沙星的浓度 ,MCPKP计算机程序处理静注及肌注的血浆药物浓度 -时间数据。健康鸡静注给药的药时数据适合二室开放模型 ,主要药物动力学参数为 :t1 /2α( 0 .1 9± 0 .0 9) h;t1 /2β( 1 .94± 0 .70 ) h;V1 ( 1 .2 2± 0 .67) L/kg;Vd(area) ( 3.89± 1 .58) L/kg;Cl B( 1 .30± 0 .1 9) L· kg- 1 ·h- 1 ;AUC( 7.89± 1 .50 ) mg· L- 1 ·h- 1 。健康鸡肌注给药的药时数据适合一级吸收二室模型 ,主要药物动力学参数为 :t1 /2 ka( 0 .2 2± 0 .1 9) h;t1 /2α( 0 .95± 0 .93) h;t1 /2β( 5.1 9±2 .0 4 ) h;tmax( 0 .67± 0 .35) h;Cmax( 1 .2 0± 0 .30 ) mg/L;AUC( 5.66± 1 .1 5) mg· L- 1 · h- 1 ;F( 71 .67±1 4 .59) %。统计矩原理处理健康鸡内服给药的血浆药物浓度 -时间数据 ,主要药物动力学参数为 :t1 /2β( 3.89±1 .1 9) h;tmax( 1 .1 8± 0 .73) h;Cmax( 0 .86± 0 .31 ) mg/L;AUC( 5.1 2± 1 .1 9) mg· L- 1 · h- 1 ;F( 59.63±1 3.8) % ;MAT( 2 .87± 1 .1 1 ) h;MRT( 5.70± 1 .1 1 ) h。沙拉沙?Pharmacokinetics of Sarafloxacin were investigated in 30 healthy Avine broiler chickens following single intravenous,intramuscular and oral administration at the dosage of 10 mg/kg b.w. of the drug.Blood samples were collected at different intervals after administration and sarafloxacin concentrations were determined by HPLC method with a fluorescent detector.The sarafloxacin concentration time data were fitted to a two compartment open model after single i.v. dosing in healthy chickens.The main pharmacokinetic parameters were as follows:t 1/2α (0 19±0 09) h;t 1/2β (1 94±0 70) h;V 1(1 22±0 67) L/kg;V d(area) (3 89±1 58) L/kg;Cl B(1 30±0 19) L·kg -1 ·h -1 ;AUC(7 89±1 50) mg·L -1 ·h -1 .The drug concentration time data were fitted to a two compartment model with first order absorption after single i.m. dosing in healthy chickens.The main pharmacokinetic parameters were as follows:t 1/2ka (0 22±0 19) h;t 1/2α (0 95±0 93) h;t 1/2β (5 19±2 04) h;t max (0 65±0 35) h;C max (1 20±0 30) mg/L;AUC(5 66±1 15) mg·L -1 ·h -1 ;F(71 67±14 59)%。After single p.o. administration in healthy chickens.The drug concentration time data were analyzed by statistical moment methods.The main pharmacokinetic parameters were as follows:t 1/2β (3 89±1 19) h;t max (1 18±0 73) h;C max (0 86±0 31) mg/L;AUC(5 12±1 19) mg·L -1 ·h -1 ;F(59 63±13 8)%;MAT(2 87±1 11) h;MRT(5 70±1 11) h.The results showed that Sarafloxacin was absorbed fast and distributed extensively in chickens.The absorption of the drug was uncomplete after single i.m. and p.o. administration and eliminated slowly after i.m. administration in healthy chickens.
