VKORC1基因1639 G>A位点多态性与新疆地区哈萨克族、汉族心房颤动患者的相关性研究

摘　　要：目的探讨新疆地区哈萨克族、汉族心房颤动(atrial fibrillation,AF)患者与维生素K环氧化物还原酶复合体亚单位1基因(vitamin K epoxide reductase complex subunit 1,VKORC1)1639G>A位点多态性的关系。方法以哈萨克族、汉族原发性心房颤动患者各100例作为病例组,以哈萨克族、汉族无心房颤动患者各100例作为对照组,从外周静脉血中提取基因组DNA,应用多聚酶链反应-限制性内切酶长度片段多态性(PCR-RFLP)方法检测VKORC1基因1639G>A位点基因型及等位基因频率。结果哈萨克族病例组和对照组VKORC1基因1639G>A位点基因型频率和等位基因频率比较差异有统计学意义(P<0.05)。汉族病例组与对照组VKORC1基因1639G>A位点基因型频率和等位基因频率比较差异有统计学意义(P<0.05)。哈萨克族病例组和汉族病例组VKORC1基因1639G>A位点基因型频率和等位基因频率比较差异有统计学意义(P<0.05)。男性病例组与女性病例组VKORC1基因1639G>A位点基因型频率和等位基因频率比较,差异无统计学意义(P>0.05)。结论 VKORC1基因1639G>A位点在新疆地区哈萨克族、汉族心房颤动患者分布中存在民族差异。Objective To investigate the association between VKORC1 gene 1639 G^A polymorphism and Atrial Fibrillation in Xinjiang Kazaks and Hansr patients. Methods Genetic DNA of peripheral venous blood was extracted from 200 primary Atrial Fibrillation patients including 100 patients from Kazakh and 100 from Han Ethnic group and 200 controls including 1O0 from Kazakh and 100 from Han Ethnic group. The 1639 G）A polymorphism of VKORC1 gene was detected by polymerase chain reaction （PCR）, and the VKORC1 gene 1639 G）A polymorphism of Chymase gene was detected using PCR restriction frag ment length polymorphism （RFLP）. Results There was difference between Kazakh AF group and Kazakh control group in VKORC1 Gene 1639 G^A polymorphism genotype frequency and allele frequency （P % 0.05）. There was also difference between Han AF group and Han control group in VKORC1 gene 1639 G 〉A polymorphism genotype frequency and allele frequency （P %0.05）. There was difference between Ka zakh Ethnic and Han Ethnic in VKORC1 Gene 1639 G^A polymorphic loci （P %0.05）. There was no difference between male AF and female AF in VKORC1 gene 1639 G^A polymorphism genotype frequen cy and allele frequency （P ~0.05）. Conclusions There is ethnic difference between Kazakh and Han in VKORC1 gene 1639 G〉A polymorphic site.
