Ge-132诱导鸡体产生外源性IFN的研究

摘　　要：以Ge - 1 32为诱生剂诱生鸡体产生外源性干扰素。将鸡胚成纤维细胞 -水泡性口炎病毒作为鸡干扰素 (IFN)的检测系统 ,对不同种细胞、不同的细胞浓度、不同的IFN诱生剂等诸因素对IFN产生的影响进行了比较。试验结果表明 :(1 )在Ge- 1 32、鸡新城疫病毒 (NDV -F)、植物血凝素 (PHA)、聚肌胞 (polyI:C)四种诱生剂中 ,诱生能力差异极显著(P <0 .0 1 ) ,且以Ge- 1 32诱生IFN的能力为最强 ,其他依次为NDV -F、PolyI :C、PHA ;(2 )鸡脾细胞和鸡白细胞产生的IFN效价高于鸡胚成纤维细胞 ;(3)IFN诱生剂最佳诱生剂量 :鸡NDV -F为 1 2 8HAU/mL ;Ge - 1 32为 70 μg/mL ;PolyI:C为 5 0 μg/mL ;PHA为 40We study exogenous interferons induced by Ge-132 in chicken.The influences of different cell species,cell consistencies as well as interferon inducer on the production of interferon were compared by the chicken fibroblast-vesicular stomatitis virus system for assay of chicken interferon.The results showed that(1) the inducing ability of the four species of interferon inducer(Ge-132,NDV-F,PHA and Poly I:C)tested had great significant difference (p<0.01) and Ge-132 possessed the strongest interferon-inducing ability,the other were NDV-F,Poly I:C,PHA in propeer order;(2)the title of chicken spleen cells and chicken leukocytes interferon was higher than that of chicken fibroblast cells;(3) the best inducing-dosage of NDV-F,Ge-132,Poly I:C and PHA was 128 HAU/mL,70 μg/mL,50 μg/mL and 40 μg/mL respectively.
