摘  要:用卵蛋白雾化吸入致敏法 ,复制豚鼠哮喘模型 ,采用密度梯度细胞分离法、双抗体夹心ELISA法、荧光酶标法和放射碱基结合分析等方法 ,设立正常组、哮喘模型组、咳喘落治疗组和地塞米松对照组 ,分析各组豚鼠肺泡灌洗液及血浆中嗜酸性粒细胞数、低密度嗜酸性粒细胞百分比、IL -4、IL -5、可溶性细胞间粘附分子 -1(sICAM -1)、嗜酸性粒细胞阳离子蛋白 (ECP)以及外周血白细胞糖皮质激素受体(GR)等指标的变化。结果显示 :咳喘落对以上指标均有明显的改善作用。提示以温阳、化痰、祛瘀法而组方的咳喘落治疗哮喘是通过影响与气道变应性炎症相关的细胞、细胞因子、粘附分子及激素受体等而发挥多环节、多靶位的免疫调控作用。In the guinea pig asthma model created by egg albumin aerosol inhalation,density gradient cell separation,double antibody, ELISA method,fluorescent enzyme labeling and radiation base conjugated analysis were applied,in the normal group,asthma model group,treatment group with “Cough and Wheezing Reliever” and control group with dexamethasone,to analyze changes in the indexes of eosinophil count in lavage solution of pulmonary alveoli and plasma,low density eosinophil percentage,IL-4,IL-5,soluble intercellular adhesive molecule 1(sICAM-1),eosinophil cationic protein(ECP)content and glucocorticoid hormone receptor(GR)of peripheral leukocyte in various groups of the guinea pigs.The results showed that “Cough and Wheezing Reliever” possesses obvious improving function for the above indexes,indicating that “Cough and Wheezing Reliever” designed upon the methods to warm yang,disperse phlegm,and dissipate stagnation acts its immune regulatory and controlling function in multiple links and targets by influencing cells,cellular factors,adhesive molecule and hormone receptor related to allergic inflammation of the air passage.
