摘 要:菌株TN9由耐受高碳氮负荷的好氧颗粒污泥中筛选获得,对其进行了初步鉴定、脱氮功能基因及胞外多聚物(EPS)方面的研究.根据16S rDNA序列确定菌株TN9的分类地位;通过PCR检测其脱氮相关基因并测序;采用高速离心-乙醇低温沉淀法提取EPS,分别测定了蛋白质和多糖的含量.结果表明:TN9为陶厄氏菌属(Thauera)细菌.检测获得了nirS、nosZ基因序列,其中该菌株的nirS与Thauera sp.27的相似性为91%,nosZ与Thauera sp.MZ1T的相似性为95%.但是16S rDNA和nirS基因、16S rDNA和nosZ基因之间的系统进化相关性较小.该菌株能够产生EPS,72 h培养物中EPS的蛋白质含量为44.4-50.7 mg/g(TS),多糖含量为16.8-18.6 mg/g(TS),蛋白质和多糖的含量比为2.4-3.0.因此,菌株TN9是一株具有脱氮和EPS分泌能力的陶厄氏菌,在废水处理方面具有应用潜力.Strain TN9 was isolated from the aerobic granular sludge tolerant to high carbon and nitrogen loads and identified by 16S rDNA gene analysis. Its nitrifying and denitrifying genes were amplified by PCR and sequenced. Extracellular polymeric substances (EPS) were extracted by high-speed centrifugation and ethanol precipitation at low temperature, and the content of protein and polysaccharide in EPS were determined. The results showed that TN9 belonged to the genus Thauera and was named Thauera sp. strain TN9. Both the nirS and nosZ gene amplified from strain TN9 had the highest similarity with Thauera strains. The similarity of nirS between TN9 and Thauera sp. 27 was 91%, and that of nosZ between TN9 and Thauera sp. MZ1T was 95%. However, low correlations were found between the phylogeny of 16S rDNA and nirS genes (R2=0.305), and between that of 16S rDNA and nosZ genes. Strain TN9 had the ability to secrete EPS. After incubation for 72 h, the protein content in EPS was 44.4-50.7 mg/g(TS), while the polysaccharide content was 16.8-18.6 mg/g(TS). The ratio of protein to polysaccharide ranged between 2.4 and 3.0. Accordingly, Thauera sp. TN9 was considered to have potential in wastewater treatment. Fig 4, Tab 1, Ref 28
