摘 要:目的:从细胞生物学水平研究运脾和络颗粒对高糖诱导的大鼠雪旺细胞增殖及NGF基因表达的影响,探讨其在糖尿病周围神经病变中防治中的意义。方法:将常规体外培养的大鼠雪旺细胞分为空白组、模型组、运脾和络颗粒低、中、高剂量组、甲钴胺组(西药对照组)、糖脉康组(中药对照组),采用MTT比色法检测高糖环境下各组雪旺细胞的增殖活性,并通过RT-PCR技术检测雪旺细胞表达NGF mRNA的能力。结果:高糖培养条件下,雪旺细胞增殖及表达NGF的能力均明显减弱,各治疗组均能对抗高糖诱导的抑增殖作用,其中运脾和络颗粒组的作用优于甲钴胺组和糖脉康组,且呈一定的时效、量效关系。结论:运脾和络颗粒能有效改善高糖诱导的雪旺细胞的逆增殖效应,并上调雪旺细胞表达NGF mRNA。Objective:To study the influence of Yunpi Heluo Granule on the proliferation of rat Schwann cells cultured in high glucose and expression of NGF mRNA.Methods:Schwann cells in culture and in vitro were divided into a blank group,model groups,Yunpi Heluo Granule groups(low,medium and high doses),mecobalamin group(control group of WM),Tangmaikang group(control group of traditional Chinese medicine).Tetrazolium salt(MTT)was used to test different groups of Schwann cell cultured in vitro;retrovirus polymerase chain reaction technology(RT-PCR)was used to test the drugs effects on rat Schwann cells NGF-mRNA Expression.Results:High glucose inhibited the proliferation and expression of NGF of SC remarkably.Each treatment group could raise expression which was inhibited by high glucose.The group of Yunpi Heluo Granule was better than the mecobalamin group and Tangmaikang group,and there were differences in time-effect and dose-effect.Conclusion:Yunpi Heluo Granulcan can restrain the inverse of the cell proliferation results under high sugar environment,and promote the expression of NGF mRNA.
