摘  要:目的:采用复方地芬诺酯灌胃建立慢性便秘大鼠模型,观察便塞通合剂对慢性便秘模型大鼠AQP3表达的影响。方法:健康SD大鼠60只,随机分为5组,空白对照组、模型组、麻仁丸治疗组、莫沙比利治疗组、便塞通合剂治疗组。模型组及各治疗组采用配制好的含复方地芬诺酯10 mg/kg混悬液4 mL灌胃,建立大鼠慢性便秘模型。正常对照组则以等量生理盐水替代复方地芬诺酯进行灌胃。模型建立后,治疗组分别以麻仁丸、莫沙比利和便塞通合剂灌胃治疗。治疗结束后,采用免疫组织化学SP法检测正常对照组、模型组及治疗各组AQP3在结肠的分布和表达情况,应用彩色病理图像分析系统对阳性表达的平均光密度值(MD)进行半定量比较分析。结果:①发现在正常对照组和慢传输便秘模型组大鼠的结肠黏膜层均有AQP3表达;②模型组AQP3平均光密度值高于空白对照组,两组比较有显著差异(P<0.05)。③与模型组相比,便塞通合剂治疗组能能抑制AQP3在大鼠结肠黏膜层的表达(P<0.05)。结论:便塞通合剂能抑制AQP3在大鼠结肠黏膜层的表达,其表达降低可能导致结肠对水分的吸收减少和(或)肠液分泌增加,从而治疗慢性便秘。Objective:To observe the effect of Biansetong Mixture on expression of Aquaporin-3 in colon tissues in the chronic constipation model rats.Methods:60 rats with chronic constipation by irrigating the rats' stomach with diphenoxylate were made,and then divided into 5 groups:control group,model group,Biansetong Mixture group,Maren Pill group and mosapride group at random.Control group was irrigated with 0.9% NaCl.Model group and treatment group were used to choose the fittest dosage of diphenoxylate(10 mg/kg)by irrigating the rats' stomach to establish the chronic constipation model.After the establishment of the model,the treatment group were respectively given Biansetong Mixture,Maren Pill and mosapride.We compared Aquaporin-3 expression by the immunohistochemistry staining in colon tissues in each group.The staining results were analyzed semi-quantitively by computerized color image analyzer using the following parameters:mean density(MD).Result:Many Aquaporin-3 stained cells were found in mucous layer of colon tissues in control group and model group.Positive cell number in tissues of the model group was higher than that in control group(P0.05).Biansetong Mixture could obviously inhibit the expression of AQP3 in colon(P0.05).Conclusions:Biansetong Mixture can inhibit the expression of AQP3 in rats with chronic constipation of colon tissues.It may be correlated to decreasing the water absorption and increasing the secretion of intestinal juice.
