摘 要:目的:观察德都红花-7味散对CCl4所致慢性肝损伤大鼠肝组织SOD,MDA及肝线粒体钠钾ATP酶活性的影响。方法:将wistar雄性大鼠,按体重随机分为空白对照组、模型组、秋水仙碱阳性对照组、德都红花-7味散低、高剂量组,共5组,每组12只大鼠。除空白对照组,其它组均腹腔注射30%CCI4橄榄油溶液2.0 mL/kg,2次/周,连续造模7周。空白对照组和模型组灌胃蒸馏水,秋水仙碱组以0.4 mg/kg,德都红花-7味散低剂、高剂量组分别以0.62 g/k g,1.04 g/kg,按2 mL/100 g,每日分2次,连续灌胃7周。末次给药后48h,10%水合氯醛0.3 mL/kg麻醉,放尽血液。迅速取出肝脏,肝左叶制备肝线粒体,肝组织匀浆,检测肝线粒体Na+-K+-ATP酶活性和肝组织匀浆SOD,MDA含量;肝右叶采用HE、Gomori、Masson方法染色,进行疗效评价。结果与空白对照组比较模型组SOD、Na+-K+-ATP酶活性明显下降,MDA含量升高,秋水仙碱组Na+-K+-ATP酶活性明显下降,有显著性差异P<0.05;与模型组比较秋水仙碱组、德都红花-7味散低、高剂量组肝组织SOD活性明显升高,MDA含量明显降低,德都红花-7味散低、高剂量组肝线粒体Na+-K+-ATP酶活性明显升高,有显著性差异P<0.05。炎症活动度及纤维化程度半定量值与模型组比较德都红花-7味散高、低剂量组炎症活动度及纤维化程度半定量值明显降低,有显著性差异,P<0.05。结论:德都红花-7味散能够明显改善CCI4引起慢性肝损伤组织病理改变,其机理可能与抗脂质过氧化,提高Na+-K+-ATP酶活性,改善肝线粒体钠泵功能有关。Objective:To observe the effects of Mongolian Medicine Deduhonghua-7 powder on SOD,MDA and Na+-K+-ATPase activity of rat with CCl4-induced chronic liver injury.Method:The male Wistar rats were randomly divided into blank control group,model group,colchicine positive control group,Deduhonghua-7 powder low and high dose groups based on body weight,12 rats in each group.Except the blank control group,other groups were injected intraperitoneally 30% of CCl4 olive oil solution at a dose of 2.0 mL/kg,twice a week,continued for 7 weeks.Blank control group and model group were given distilled water orally;colchicine group was given 0.4 g/kg colchicine,Deduhonghua-7 powder low-dose group and high-dose group were given Deduhonghua-7 powder at doses of 0.62 g/kg,1.04 g/kg respectively,2 mL/100 g,twice a day,continued for 7 weeks.48 hours after last administration,the rats were fasted 12 hours,then anesthetized by 10% of chloral hydrate 0.3 mL/kg,the blood was exhausted at last.The liver and left lobe of liver were removed immediately for preparing the liver mitochondria and liver homogenates so as to detect the Na+,K+ ATPase activity and SOD and MDA contents of liver homogenates.The HE,Gomori and Masson method were used to stain the right lobe of liver and evaluate the efficacy.Result:Compared with blank control group,the SOD and Na+-K+-ATPase activity decreased significantly,the content of MDA was increased.The Na+-K+-ATPase activity of colchicine group was significantly decreased with significantly differences(P0.05).Compared with model group,SOD activities of colchicine group,Deduhonghua-7 powder low and high dose group were significantly increased,and MDA content was significantly decreased.Na+-K+-ATPase activities of Deduhonghua-7 powder low and high dose group were significantly increased with significantly differences(P0.05).Compared with model group,semi-quantitative value of the inflammatory activity and fibrosis of Deduhonghua-7 powder high and low dose groups were significantly d
