大肠癌辅助化疗期血瘀证血清蛋白质组学探究

摘　　要：目的:筛选大肠癌辅助化疗期血瘀证血清差异表达的蛋白质。方法:收集大肠癌辅助化疗期血瘀证、脾虚证及平和体质患者外周血清各8例,通过双向凝胶电泳以及质谱鉴定和生物信息学分析,寻找在大肠癌辅助治疗期血瘀证患者血清中特异表达的蛋白质。结果:对表达差异明显的11个蛋白质点进行质谱鉴定和生物信息学分析发现,在瘀血证及脾虚证大肠癌患者中转移相关蛋白、触珠蛋白、微管蛋白α链3、Rab GDP分解抑制剂β、血清载脂蛋白A-I均高表达。凝血酶原、二氢叶酸还原酶样蛋白质、血管生长因子受体-1、肿瘤蛋白DJ-1在血瘀证中高表达但在脾虚证中低表达,神经生长因子G-1、白介素27α在血瘀证患者中低表达而在脾虚患者中高表达。结论:11种蛋白差异表达蛋白质的组合性变化可能是大肠癌血瘀证的物质基础,具有潜在的诊断、预后标志物或治疗靶点的意义。Objective：To seek differentially expressed serum proteins of colorectal cancer patients with blood stasis syndrome of TCM in colorectal cancer in adjuvant chemotherapy stage.Methods：Two-dimensional difference gel electrophoresis and mass spectrometry were used to screen the differentially expressed plasma proteins among 24 patients with blood stasis syndrome,syndrome of spleen deficiency or constitution of yin-yang harmony.Results：Metastasis-associated protein MTA1,Haptoglobin,Tubulin alpha chain-like 3（TUBAL3）,Rab-GDP-dissociation inhibitor beta and Apolipoprotein A-I were highly expressed in the serum of the patients with blood stasis syndrome and syndrome of spleen deficiency.Prothrombin,Dihydrofolate reductase-like protein 1,Vascular endothelial growth factor receptor 1 and Protein DJ-1 were only highly expressed in the patients with blood stasis syndrome while decreased in syndrome of spleen deficiency.Netrin-G1,Interleukin-27 subunit alpha were decreased in the patients with blood stasis syndrome while were highly expressed in syndrome of spleen deficiency.Conclusion：The proteins identified in this study may be related to the development of blood stasis syndrome in patients with colorectal cancer and represent useful molecular targets for diagnosis and treatment of the syndrome.
