小型动物肺脏灌流固定装置的研制和应用

摘　　要：目的建立控制简便、稳定且可规范化操作的适合小型实验动物肺脏灌注固定装置。方法该装置包括用于放置动物肺脏的工作台、用于盛放固定液的灌流容器、灌流管、带刻度的支撑杆和用于固定灌流容器的固定夹等，能有效实施小动物肺脏的灌注固定。应用该肺脏灌流固定装置分别对大鼠及小鼠肺脏进行灌注固定，石蜡包埋后进行切片，HE染色，与常规直接固定方法进行比较。结果该小型动物肺脏灌流固定装置可快速、便捷、标准化地对小鼠、大鼠肺脏进行固定，并且灌注固定的肺脏石蜡包埋切片后HE染色，肺脏组织、小血管及气管组织结构清晰，背景干净。结论研制的小型动物肺脏灌流固定装置应用方便，调节灵活，结构简单，成本低，易操作，能对小型动物肺脏有效进行固定，经灌注固定肺组织切片明显优于直接固定法。Objective To establish an easy, stable, and standardized operating system specifically designed for small animal lung perfusing experiments. Methods This new designed set of system contains workbench for placing the animal lungs, container for blooming the fixing solution during perfusion, perfusing tubes, scale labeled supporting bar, and clips for holding the perfusing container. It is highly effective to perform small animal lung perfusing with this system. The lungs of both rats and mice were perfused by this new designed system and the regular set, the perfused lungs was embedded with paraffin and sectioned. HE staining was done and comparison was performed between these two perfusing systems. Results This new designed system brought faster, more convenient and standardized perfusing of lungs of rats or mice, compared to the regular perfusing system. Moreover, HE staining results indicated that the structure of the lung tissue, small blood vessels, and trachea tissue was clearer in section perfused with the new designed system than that with the regular perfusing system, simultaneously, a cleaner background was also revealed as well. Conclusions The new designed perfusing system is easy to operate, convenient to adjust, simply designed with low expense. It is highly effective to perfuse lungs of small animals. Results from paraffin embedded sections suggest that it is significantly better than the regular perfusing set.
