摘  要:目的探讨辛开苦降方干预对初发2型糖尿病(T2DM)KKay小鼠肝胰岛素抵抗及脂质代谢的影响。方法将T2DMKKay小鼠按血糖轻重程度随机分为辛开组、苦降组、辛开苦降组、马来酸罗格列酮组和模型组。选取C57BL/6J小鼠10只作为正常对照组(正常组)。各组连续灌胃给药4周后取材,测定其空腹血糖、血清胰岛素、血脂、肝糖原、肝脂质含量,显微镜下观察肝脏病理形态。结果给药4周后,与模型组比较,苦降组、马来酸罗格列酮组血糖明显降低(P<0.05),而辛开组、辛开苦降组血糖差异无统计学意义(P>0.05)。与模型组比较,各给药组小鼠胰岛素浓度均降低,胰岛素敏感指数均升高(P<0.05),但各给药组组间差异无统计学意义(P>0.05)。与模型组比较,马来酸罗格列酮组CHO、TG、LDL值均降低,苦降组、辛开苦降组TG值降低,差异均有统计学意义(P<0.05)。与模型组比较,辛开苦降组肝TG含量降低(P>0.05)。肝脏病理显示:给药4周后,模型组小鼠肝脏结构紊乱,肝细胞胞浆有明显的空泡,肝细胞脂肪变性;各给药组小鼠肝脏结构较模型组完整,肝细胞脂肪变性较模型组轻。结论苦降方可明显降低T2DM KKay小鼠血糖,辛开苦降方可降低血清胰岛素浓度,增强胰岛素敏感性,升高肝糖原含量,降低血脂和肝脂质中TG水平,进而减轻肝胰岛素抵抗,改善T2DM脂代谢紊乱状态。Objective To explore the effect of acrid-opening and bitter-downbearing prescription intervention on insulin resistance and lipid metabolism in KKay mice with early-satge type 2 diabetes mellitus (T2DM). Methods KKay mice with T2DM were divided into an acrid-opening group (Group A), a bitter-downbearing group (Group B), an acrid-opening and bitter-downbearing group (Group C), a Rosigiitazone maleate group (Group R) and a model group (Group M) according to blood sugar level; C57BL/tJ mice (n=10) were selected as a normal control group (Group N). Materials were taken after 4-week continuous intragastrie administration in each group, contents of fasting blood-glucose, serum insulin, blood lipid, hepatic glycogen and liver lipid were detected, and pathological morphology in liver was observed under the microscope. Results Compared with the Group M, the level of blood sugar in the Group B and R was apparently decreased (P 〈 0.05), while that in the Group A and C was statistical insignificance (P 〉 0.05). Compared with the Group M, the insulin concentration of mice in the treated groups was decreased and the insulin sensitivity index was increased (P 〈 0.05), however, there were no significant differences among the treated groups(P 〉 0.05). Compared with the Group M, the levels of cholesterol(CHO), triglyceride (TG) and low-density lipoprotein (LDL) in the Group R as well as TG in the Group B and C were decreased with significant differences (P 〈 0.05). Compared with the Group M, the level of liver TG in the Group C was decreased(P 〉 0.05). According to hepatic pathology, for the Group M, the structure of liver was disorganized and there were clear vacuoles in the cytoplasm of hepatic cells and fatty degeneration in hepatic cells; for the treated groups, the structures of liver were relatively intact and the fatty degeneration of hepatic cells was lighter than that in the Group M.
