摘  要:目的探讨围生期暴露双酚A(bispheno1 A,BPA)对母鼠及其成年期雌性仔鼠氧化应激的影响。方法将21只清洁级SD妊娠大鼠分为3组,分别为对照(含1%无水乙醇的水)组和低(1μg/ml)、高(10μg/ml)浓度BPA组,每组7只。从妊娠第6天至哺乳期(出生后20 d)结束,母鼠采用自由饮水方式进行染毒;断乳后,雌性仔鼠采用自由饮水方式继续喂养80 d。测定断乳后母鼠和100日龄雌性仔鼠血清丙二醛(MDA)含量和超氧化物歧化酶(SOD)活力及总抗氧化能力(T-AOC)。结果与对照组比较,断乳后各浓度BPA组母鼠血清MDA含量均增加,SOD活力和T-AOC水平均降低,差异有统计学意义(P<0.05);高剂量BPA组母鼠血清MDA含量高于低剂量BPA组,差异有统计学意义(P<0.05),且随着BPA暴露浓度的升高,断乳后母鼠血清MDA含量呈上升趋势,SOD活力和T-AOC水平均呈下降趋势。与对照组比较,出生后100 d时各浓度BPA组雌性仔鼠血清MDA含量均增加,仅高浓度BPA组雌性仔鼠血清SOD活力和T-AOC水平均降低,差异有统计学意义(P<0.01);且随着BPA暴露浓度的升高,出生后100 d雌性仔鼠血清SOD活力和T-AOC水平均呈下降趋势。结论围生期暴露BPA可引起母鼠和成年期雌性仔鼠氧化应激的发生。Objective To explore the effect of perinatal exposure to bisphenol A (BPA) on the parameters of oxidative stress in maternal and adult female offspring rats. Methods A total of 21 pregnant Sprague-Dawley rats were randomly divided into three groups,seven rats in each group, the control group (1% of anhydrous alcohol with water),low dose (1 μg/ml) and high dose (10 μg/ml) groups of BPA exposed through drinking water,from gestation day 6 to the end of lactation respectively. The serum malondialdehyde (MDA),superoxide dismutase (SOD) and total antioxidant capacity (T-AOC) were measured in maternal after weaning and female offspring after postnatal day 100. Results Compared with the controls,the serum MDA level significantly increased,but serum SOD activity and T-AOC levels were significantly decreased in maternal rats of lower and higher BPA groups (P〈0.05). There was an increasing trend in maternal serum MDA levels with the increasing BPA doses. The serum MDA level was significantly higher in offspring of lower and higher BPA groups, and serum SOD activity and T-AOC levels were significantly lower in offspring of higher BPA group than in the control (P〈0.05). There was a decreasing trend in SOD activity and T-AOC levels with the increasing BPA doses. Conclusion Perinatal exposure to BPA may induce oxidative stress in maternal and adult female offspring rats.
