摘 要:目的探讨不同剂量亚硒酸钠对氯化镉诱导的雄性SD大鼠肝细胞的凋亡率、c-Myc和P53蛋白表达的影响。方法将65只健康SPF级SD雄性大鼠随机分为13组,分别设对照(生理盐水)组、氯化镉(5.0、10.0、20.0μmol/kg)组及亚硒酸钠(2.5、5.0、10.0μmol/kg)+氯化镉(5.0、10.0、20.0μmol/kg)联合作用组,每组5只。氯化镉采用一次性腹腔注射染毒,亚硒酸钠采用一次性灌胃染毒。染毒后48 h,用流式细胞仪检测细胞凋亡率,免疫组化法测定c-Myc和P53蛋白的表达量。结果与相同剂量氯化镉组相比,亚硒酸钠剂量为2.5μmol/kg时,全部联合作用组的大鼠肝细胞凋亡率、c-Myc和P53蛋白表达均降低(P<0.05或P<0.01);而亚硒酸钠剂量为5.0、10.0μmol/kg时,部分联合作用组的大鼠肝细胞凋亡率、c-Myc和P53蛋白表达降低(P<0.05或P<0.01)。大鼠细胞凋亡率与c-Myc、P53蛋白表达量均呈正相关(P<0.01)。结论2.5μmol/kg的亚硒酸钠对氯化镉所诱导的大鼠肝细胞的凋亡及c-Myc和P53蛋白表达的升高具有一定的拮抗作用。Objective To study the effects of sodium selenite at diverse dose levels on apoptosis and the expression of c- Myc and P53 protein induced by CdC12 in male Sprague-Dawley rat hepatocytes. Methods Sodium selenite at the doses of 2.5,5.0 and 10.0 μmol/kg combined with cadmium at the doses of 5.0,10.0 and 20.0 μmol/kg cadmium chloride was used pairwise. Cadmium and selenium were administrated intraperitoneally and through gavage respectively. In control group, the rats received no selenium and cadmium treatment. All treatments were conducted only one time. 48 h after the initial treatment. apoptosis was detected with flow cytometry,c-Myc and P53 proteins were determined by immm:ochemistry, in rats' hepatocytes. Results In all combined treatment groups with sodium selenite at the doses of 2.5 μ mol/kg, the levels of apoptosis rate, c-Myc and P53 proteins were lower significantly than those in corresponding CdC12 group (P〈0.05 or P〈0.01). However,just in some combined treatment groups with sodium selenite at dose of 5.0 and 10.0μmol/kg,apoptosis rate,c-Mye and P53 protein expression induced by CdC12 were lower (P〈0.05 or P〈0.01). The Pearson correlation coefficient between c-Myc,P53 expression and apoptosis were 0.975 and 0.943 respectively (P〈0.01). Conclusion Selenium only at dose of 2.5 μmol/kg may inhibit apoptosis induced by cadmium in rat hepatocytes.
