摘 要:为了分析和田羊毛囊角蛋白关联蛋白(KAP)1.1基因,试验以南疆地方品种羊(和田羊)为样本,从和田羊毛囊中提取总RNA,设计1对引物,通过PCR反应扩增出长度为535 bp的基因片段,连接到pMD18-T载体上。结果表明:克隆得到和田羊毛囊KAP1.1基因成熟蛋白编码序列,序列长度与GenBank上发表的序列长度相同,经生物信息学软件分析,有2处碱基发生突变,同源性为99%,说明试验成功克隆了KAP1.1基因。To analyze the keratin- associated protein (KAP) 1.1 gene of the hair follicles in Hetian sheep, which was the local breed sheep in south region of Xinjiang, the Hetian sheep as a sample were selected to extract the total RNA from the hair follicles. A pair of primers was de- signed, and a 535 bp gene fragment was amplified by PCR, and then was connected into the pMD18 -T vector. The results showed that the se- quence of KAPI. 1 gene encoding mature protein in the hair follicles in Hetian Sheep was obtained, the length of its sequence was same as that of the sequence published on GenBank. Bioinformatics software analysis showed that there were two base mutations, and the homology was 99%. The results indicate that KAP1. 1 gene is cloned successfully.
