摘  要:本研究合成了三聚氰胺半抗原及完全抗原并鉴定合成是否成功,为制备三聚氰胺抗体奠定基础。将2-氯-4,6-二氨基-1,3,5-三嗪和3-巯基丙酸的反应合成半抗原3-(4,6-二氨基-1,3,5-三嗪-2-基硫代)丙酸,采用质谱鉴定和红外光谱鉴定;通过活性酯法将半抗原分别与牛血清白蛋白、卵清蛋白进行偶联制备完全抗原,采用紫外光谱法初步鉴定,后经动物免疫进一步验证。结果表明半抗原与完全抗原合成成功,为后续的抗体制备奠定了基础。To synthesize and identify the hapten and complete antigens of melamine for the purpose of preparation of anti-melamine antibodies,2-chloro-4,6-diamino-1,3,5-triazine were reacted with 3-mercaptopropionic acid to produce hapten (3-(4, 6-diamino-1,6-dihydro-1,3,5-triazin-2-ylthio) propanoic acid). The molecular structure of the hapten was identified by mass spectrometry and infrared spectrometry. The hapten was respectively conjugated to carrier protein bovine serum albumin and egg ovalbumin to generate complete antigens by the active ester method and the conjugates were preliminarily identified by ultraviolet spectrum and further validated by animal immunization. The results indicated that the hapten and complete antigens were prepared successfully. Synthetic routes were feasible and the results laid the foundation for the following preparation of anti-melamine antibodies.
