补肾解毒方中马钱苷和黄芩苷含量的高效液相色谱测定

摘　　要：目的建立测定补肾解毒方中马钱苷和黄芩苷的含量的方法。方法采用HPLC,色谱柱为Kromasil ODS-1 C18柱(100mm×4.6 mm,5μm)。①黄芩苷:流动相为甲醇-0.1%磷酸溶液(42∶58),检测波长280 nm;流速1.0 ml/min;进样量5.0μl;②马钱苷:流动相为水-乙腈(85∶15),检测波长236 nm,柱温为40℃,流速1.0 ml/min,进样量10μl。结果补肾解毒方中黄芩苷在0.005～2.2μg浓度范围内线性关系良好(r=0.999 9),平均回收率为98.18%;马钱苷在0.999 6～199.92μg浓度范围内线性关系良好(r=1.0),平均回收率为99.72%。结论该方法简便、准确、分离效果好,可用于补肾解毒方的质量评价。Objective To establish the determination method of loganin and baicalin in Bushen Jiedu recipe by HPLC. Methods HPLC was used with Kromasil ODS-1 C18(5 μm,100mm×4.6 mm) as the analytical column,The flow rate was 1.0ml/min;①Baicalin:mobile phase was Methanol 0.1% Phosphate(42∶58).The UV detective wave length was at 280 nm.The sample volume was 5.0 μl.②Loganin: mobile phase was Pure water Acetonitrile(85:15),The UV detective wave length was at 236 nm.The sample volume was 10.0 μl. Results Baicalin showed good linear relationship(r=0.999 9)with in the range of 0.005~2.2 μg,and the average recovery rate was 98.18%(RSD=4.1%);Loganin showed good linear relationship(r=1.0)with in the range of 0.9996~199.92 μg,and the average recovery rate was 99.72%(RSD=2.0%). Conclusion The method is simple,accurate and durable for quantitative determination of of loganin and baicalin in Bushen Jiedu recipe.
