摘 要:目的观察泽泻含药血清对ox-LDL诱导损伤的血管内皮细胞活性、形态学、NO、NOS及SOD的影响,探讨泽泻含药血清保护血管内皮细胞的可能机理。方法 100μmol/L ox-LDL造成血管内皮细胞氧化应激损伤模型,cck-8法检测各组细胞增殖情况;试剂盒检测各组细胞上清液中SOD的活力和NOS、NO含量。结果 100μmol/L ox-LDL可抑制血管内皮细胞活性,降低SOD活力,降低NOS含量和NO的分泌,而泽泻则能改善100μmol/L ox-LDL对血管内皮细胞的损伤,对血管内皮细胞具有一定的保护作用。结论泽泻可以通过调控抗氧化损伤机制保护血管内皮细胞,这可能是泽泻抗氧化损伤机理所在。Objective To obtain the influence of the serum containing active ingredients of Rhizoma Alismatis on the viability,morphology,SOD,NOS and NO of the vascular endothelial cell damaged by ox-LDL,and to study the mechanism of anti-oxidative damage of the serum on the cells. Methods The endothelial cells treated with 100 μmol/L ox-LDL were used as an oxidative stress model.The method of cck-8 was used to detect cell proliferation,and xanthine oxidase and nitrate reductase assay were used to detect SOD activity,NOS and NO content in the medium supernatant of each group,respectively. Results In the model groups,vascular endothelial cell viability was inhibited partly and their SOD activity,NOS and NO secretion were decreased.While in the administration groups the oxidative damage of vascular endothelial cells was reduced and the active ingredients of Rhizoma Alismatis have a protective effect on oxidatively damaged vascular endothelial cells. Conclusion The mechanism of oxidative damage of the active ingredients of Rhizoma Alismatis could be that it reduced the oxidative damage of endothelial cells to improve the cell survival.
