摘  要:目的建立HPLC-ELSD测定地黄不同炮制品中低聚糖含量的方法,研究不同炮制方法对熟地黄低聚糖含量的影响。方法采用高效液相色谱法,以蒸发光散射检测器进行检测;色谱柱:prevail carbohydrate ES柱(5μm,4.6 mm×250mm);流动相:乙腈-水(65.5∶34.5);流速:0.8 ml.min-1;柱温:30℃;ELSD条件:漂移管温度60℃,空气流速:2.0 L.min-1,增益:1。结果蔗糖平均回收率为98.44%,RSD=1.70%;棉子糖平均回收率为99.57%,RSD=2.46%;水苏糖平均回收率为99.21%,RSD=2.06%;生地、炆制地黄、清蒸地黄、酒炖地黄中低聚糖含量分别为63.88%,(3.21±1.17)%,(31.06±6.83)%,(32.33±7.04)%。结论该方法操作简便,可靠稳定,结果准确;生地黄中低聚糖含量高于地黄炮制品,炆制地黄低聚糖含量低于其他两种方法,为阐明其炮制机理提供实验依据。Objective To establish an HPLC-ELSD method for determination of Oligosaccharide contents in different processed Radix Rehmannia Praeparata and to study the effect of different processed method on Oligosaccharide contents in Radix Rehmannia Praeparata. Methods HPLC-ELSD was applied to analyze the content of Oligosaccharide with prevail carbohydrate ES column(5μm,4.6 mm×250 mm),mobile phase acetonitrile-water(65.5∶34.5),flow rate 0.8ml·min-1,column temperature 30℃,temperature of drift tube 60℃,gas flow 2.0 ml·min-1 and gain 1. Results The sucrose,raffinose and stachyose average recovery was 98.44%(RSD=1.70%),99.57%(RSD=2.46%)and 99.21%(RSD=2.06%) respectively.the content of Oligosaccharide in radix rehmanniae,Wenzhi Dihuang,steaming Dihuang and stewing it by Liqour were 63.88%,(3.21±1.17)%,(31.06±6.83)%,(32.33±7.04)% respectively. Conclusion The method is simple,accurate and reproducible.The content of Oligosaccharide in Wenzhi Dihuang was lower than that of steaming Dihuang and stewing it by Liqour.It provided an experimental basis to illustrate its processing mechanisms.
