六溴环十二烷单一及与多溴联苯醚复合暴露对发育期大鼠抗氧化系统的影响  被引量:3

Effects of single exposure to HBCD and composite exposure to HBCD and PBDEs on the antioxidant system of new-born rats

摘  要:模拟真实环境中六溴环十二烷(HBCD)的暴露水平,研究HBCD单一及与商用多溴联苯醚DE-71复合暴露对发育期大鼠肝组织抗氧化系统的影响。选择单一HBCD(10μg/kg、50μg/kg、100μg/kg及300μg/kg)及HBCD与等质量比的DE-71复合试剂对新生3 d大鼠进行为期42 d的暴露。取肝组织测定谷胱甘肽过氧化物酶(GPx)、谷胱甘肽还原酶(GR)的活性,还原型谷胱甘肽(GSH)、氧化型谷胱甘肽(GSSG)及脂质过氧化物丙二醛(MDA)浓度的变化。结果表明,HBCD单一暴露时,各暴露组GSH浓度均显著升高(p<0.05),GPx、GR活性在10~100μg/kg、50~300μg/kg暴露剂量下被显著诱导(p<0.05),MDA、GSSG浓度无显著变化(p>0.05)。HBCD/DE-71复合暴露时,暴露组大鼠肝脏GSH浓度整体呈先升高后降低趋势,300μg/kg暴露组GSH浓度显著下降(p<0.05);各暴露组GPx活性无显著变化(p>0.05),但GR活性均显著降低(p<0.05);100~300μg/kg暴露组大鼠肝脏中GSSG浓度显著降低(p<0.05),同时MDA浓度显著增加(p<0.05)。研究表明,与HBCD单一暴露的影响相比,HBCD/DE-71复合暴露使大鼠体内抗氧化酶活力降低,脂质过氧化加剧,氧化损伤严重,引发的健康危害应该重视。The paper is concerned about the effects of single exposure to hexabromocyclododecane (HBCD) and the composite exposure to HBCD and polybrominates diphenyl ethers (PBDES) on the antioxidant system of rats under development through a numerical simulation research. Thus, as a matter of fact, we adopted the method of lavage to the controlled groups whereas we adopted the gavage peanut oil to the exposed groups with HBCD and HBCD/DE - 71, that is to say, the control group should be given the same volume of the pure peanuts. When testing the Sprague-Dawley 3-day new-born rats, they were let to be exposed to different doses of HBCD ( 10 μg/kg, 50 μkg, 100μg/kg, 300 μg/kg body weight) and the same concen- tration of HBCD and DE - 71 composite doses ( 10 μg/kg, 50 μg/kg, 100 μg/kg, 300 μg/kg body weight) to the rats of 42 days so as to test the activities of the glutathione peroxidase (GPx), glutathione reductase (GR) and the contents of the reduced glutathione (GSH), the oxidized glutathione (GSSG) and lipoperoxide (MDA). That is to say, our tests were given to the exposed groups with the gavage peanut oil with HBCD and HBCD/DE - 71, whereas the control groups were exposed to the same volume of pure peanut oil instead. The results of out tests show that, when exposed to HBCD, all the two groups of GSH tend to increase their activities significantly, i.e. p 〈 0.05. The activity of GPx was found to be induced noticeably in the range of exposure doses between 10- 100 μg/kg whereas the activity of GR was induced remarkably in the range of 50 - 300 pg/kg (p 〈 0.05). However, the contents of GSSG and MDA were not found statistically increased in each exposure group (p 〉 0.05). When exposed to HBCD/DE - 71, the activity of GPx in all the exposure groups were found higher than in the control group, but have not statistically, on the contrary, GR was found decreased significantly (p 〈 O. 05).
