摘 要:目的比较两种不同方式灭活的大肠杆菌(OP50)对秀丽隐杆线虫生长发育的影响,以选择合适的灭活食物菌的方法。方法分别采用热处理(65℃水浴)和钴60照射(2000 Gy)的方法灭活OP50,从秀丽隐杆线虫生长发育的影响、OP50的蛋白组成改变以及琥珀酸脱氢酶活性改变等角度评价热处理组、钴60照射组和对照组间的差异,从而确定最有利于秀丽隐杆线虫生长发育的OP50灭活处理方式。结果钴60照射OP50组饲喂后,秀丽隐杆线虫的生长发育与对照组无差异,而热处理OP50组线虫的发育显著迟缓于对照组。同时噻唑蓝[3-(4,5)-dimethylthiazol(-2-y1)-2,5-diphenyltetrazolium bromide,MTT]检测结果显示,热处理OP50的琥珀酸脱氢酶活性明显低于对照组和钴60照射组。结论钴60照射是秀丽隐杆线虫实验研究中比较合适的OP50灭活方法。钴60照射和热处理OP50对秀丽隐杆线虫生长的差异可能由于OP50细菌中蛋白活性差异所致。本文为排除药物和毒物可能会间接通过影响OP50从而影响秀丽隐杆线虫的发育过程的干扰因素、提高基于秀丽隐杆线虫研究数据的可靠性提供了重要参考。Objective To find a suilable method for preparing the inactivated Escberichia coli OP50 based on the influ-ence of Caenorhabditis elegans on the growth and development of E.coli OP50.Methods OP50 was inactivated by heat treatment(65℃)and ^60Co irradiation(2000Gy)respectively followed by the observation of the growth of C. elegans.The activity of succinate dehydrogenase was further analyzed by the MTT method to delermine the ettect ot these two operations on protein activity of OP50. Results There was no difference in the growth of C. elegans between the ^60Co irradiaton group and control group. However, the development of heat treatment group was significantly delayed compared to the control group. 3-(4,5 ) -Dimethyhhiazo ( -2-yl ) -2,5-diphenyltetrazolium bromide (MTT) results showed that after heat treatment the succinate dehydrogenase activity of OPSO was significantly lower than that of the control group and ^60Co irradiation group. Conclusion The inactivation method of ^60Co irradiation is a suitable approach to OP50 in the C.elegans experi-ment. The different effect of ^60Co irradiation and heat treatment of OP50 on C.elegans growth may be due to the deteriora-tion of barterial proteins activity. Our study provides important reference for improving the reliability of studies using C.ele-gans by eliminating the potenlial factors that might affect the growth and development of C.elegans through OP50 indirectly.
