摘 要:目的观察不同输液方案对兔肺组织水通道蛋白5(AQP5)表达的影响。方法清洁级成年雄性SD家兔54只,随机分为3组(n=18):限制性输液组(A组,输液速度5 ml/h),常规输液组(B组,输液速度10 ml/h),急性高容量血液稀释组(C组,输液速度20 ml/h)。各组动物分别于输液前(T0)、输液后30 min(T1)、输液后60 min(T2)、输液后120 min(T3)记录心率(HR)、平均动脉压(MAP)、中心静脉压(CVP)等血流动力学指标。将A组在T1、T2、T3时刻再分为3组(n=6):A0.5组(T1)、A1.0组(T2)、A2.0组(T3),B、C组再分组时刻同A组。然后,分别于T1、T2、T3时刻处死动物,取相同部位右肺组织制作HE切片,光镜观察肺组织结构变化;同时取肺组织标本于透射电镜下观察肺组织超微结构变化。免疫组化染色检测AQP5的表达,图像分析测定AQP5染色的平均灰度和阳性单位。结果光镜下各组实验动物肺组织无病理学改变;透射电镜观察各组实验动物肺组织超微结构无变化。免疫组化和图像分析结果显示,肺泡Ⅰ型上皮细胞有大量AQP5阳性染色;不同输液方式对AQP5平均灰度和阳性单位的影响差异无统计学意义(P﹥0.05)。结论不同输液方式对肺组织AQP5的表达无显著影响。Objective To observe the effect of different infusion modes on the expression of aquaporin-5 in the lung of rabbits. Methods Fifty-four healthy adult male Clean SD rabbits were randomly divided into 3 groups (n = 18) : restricted fluid administration group (group A, transfusion speed 5 ml/h) ;conventional fluid administration group (group B, l0 ml/ h) and AHH ( group C, 15 ml/h). HR, MAP, CVP were recorded before infusion (To), and 30, 60, 120 min after in-fusion (T1-3 ). Group A was divided into 3 subgroups (n =6) at T1, T2, T3 : group A0.5, group A1.0 and group A2.0. Group B and C were divided in the same way. The rabbits were sacrificed, and the same part of pulmonary tissues was taken out at T1-3. The changes of lung tissues were observed under a light microscope and transmission electron microscope. The expression of AQPs was determined by immunohistochemical staining test ,while the value of the average gray scale and posi- tive unit was determined by image analysis. Results There was no difference in lung tissues under the light microscope or transmission electron microscope between the groups. A large amount of AQP5 positive staining was found in Type I alveolar epithelial cells by immunohistochemistry and image analysis technology. No difference was found in the value of A and posi- tive unit of AQP5 in different infusion modes (P 〉 0.05 ). Conclusion Different infusion modes have no effect on the ex- pression of AQP5 in the lung of rabbits.
