PPARβ激动剂在大鼠创伤性脑损伤中的作用及机制

摘　　要：目的观察过氧化物酶体增生物激活受体β(PPARβ)激动剂GW0742对大鼠创伤性脑损伤的影响及作用机制。方法选用健康SD大鼠,随机分为单纯脑损伤组和GW0742治疗组,制备自由落体脑损伤模型,分别于损伤后1d、2d、3d、4d、5d、6d和7d进行运动功能测试和乳酸脱氢酶(LDH)活性检测;于损伤后24h通过干湿重法检测脑水含量,通过ELISA检测TNF-α和NF-κB的表达水平。结果与单纯损伤组相比,GW0742治疗组各时间点的运动功能评分均明显升高(P<0.01),而且LDH含量均明显减少(P<0.05)。损伤后24h,GW0742治疗组的脑水含量(P<0.01)及TNF-α和NF-κB表达水平(P<0.05)明显低于单纯损伤组。结论 PPARβ激动剂GW0742对大鼠创伤性脑损伤有明显的保护作用,抗炎作用可能是GW0742发挥保护作用的机制之一。Objective To study the effect of peroxisome proliferator-activated receptorsβ （PPARβ） agonist, GW0742, on traumatic brain injury in rats. Methods One Sprague-Dawley （SD） rats were randomly divided into brain injury group and GW0742-treated group, and brain trauma was induced by a weigh-drop method. At 1,2,3,4,5,6 and 7 day after traumatic brain injury, motor function was measured using a wire-grip test and LDH activity was detected by spectrophotometric. Brain water content was measured by dry and wet weights of brain, and the contents of TNF-ct and NF-κB in brain tissues were detected by ELISA at 24h after traumatic brain injury. Results Compared with injury group rats, wire grip scores were significantly higher （ P 〈 0.01 ） and LDH activity was significantly reduced （ P 〈 0.05 ） at each time point after traumatic brain injury in GW0742-treated group rats. Brain water content （ P 〈 0.01 ） and the expression of TNF-α and NF-κB in brain tissues （P 〈 0.05 ） in GW0742-treated group rats were lower than in injury group rats at 24h after traumatic brain injury. Conclusion GW0742, a PPAR beta agonist, has a neuroprotective effect on traumatic brain injury in rats, and the anti-inflammatory effect of GW0742 may contribute to a beneficial effect against traumatic brain injury.
