摘  要:目的:研究羟基红花黄色素A(HYSA)对抗原诱导大鼠嗜碱性细胞白血病细胞株(Rat basophilic leukemiacell line,RBL-2H3)细胞脱颗粒的影响。方法:采用MTT法检测HYSA对RBL-2H3细胞增殖活性的影响;每孔加入抗DNP-IgE单抗(0.25μg/ml)与二硝基苯基人血清白蛋白(30ng/ml)以复制抗原诱导细胞脱颗粒模型,研究HYSA对抗原诱导RBL-2H3细胞活化脱颗粒的抑制作用;采用Westernblot法检测HYSA对抗原诱导RBL-2H3细胞活化脱颗粒丝裂原活化蛋白激酶(MAPK)蛋白表达的影响。结果:3~300μmol/L的HYSA对RBL-2H3细胞的增殖无明显影响;300μmol/LHYSA能显著降低RBL-2H3细胞脱颗粒程度以及抑制MAPK蛋白的表达。结论:HYSA对抗原诱导RBL-2H3细胞活化脱颗粒的炎症反应具有一定的抑制作用,且其作用可能与抑制MAPK蛋白表达有关。OBJECTIVE: To study the effects of Hydroxysaffior yellow A (HYSA) on antigen-stimulated degranulation in RBL-2H3 cells. METHODS: MTT assay was performed to establish the effect of HYSA on the RBL-2H3 cells viability; the RBL-2H3 cells were stimulated with 0.25 μg/ml anti-DNP IgE and 30 ng/ml DNP-HSA to establish degranulation model, the inhibitory effect of HYSA on antigen-stimulated degranulation in RBL-2H3 cells was researched; MAPK protein expression after antigen-induced degranulation was examined by the Western blot analysis in RBL-2H3 cells. RESULTS: HYSA(3-300 μmol/L)had no significant effect on RBL-2H3 cells proliferation. HYSA (300 μmol/L)significantly decreased the degranulation and MAPK phosphorylated protein level in RBL-2H3 cells. CONCLUSIONS:HYSA inhibited the inflammation on antigen-stimulated degranulation in RBL-2H3 ceils through inhibiting of the MAPK protein phosphorylation.
