摘 要:目的:研究天麻素对Ox-LDL诱导牛主动脉内皮细胞(BAECs)黏附HL-60细胞的抑制作用及其机制。方法:通过细胞黏附实验观察天麻素对BAECs与HL-60细胞的黏附作用;流式细胞术检测ICAM-1和VCAM-1的表达;细胞色素C还原法测定细胞含量;报告基因法检测核转录因子κB(NF-κB)转录活性的变化。结果:天麻素可使Ox-LDL诱导的BAECs黏附HL-60细胞显著降低,BAECs细胞表面ICAM-1和VCAM-1表达下降,细胞含量以及NF-κB转录活性也显著下降。结论:天麻素能抑制Ox-LDL诱导的BAECs黏附HL-60细胞。Objective: To investigate the inhibitory effect of gastrodine on Ox-LDL-stimulated adhesion of HL-60 cells to bovine artery endothelial cells (BAECs) and the underlying mechanism. Methods: Cell adhesion assay was used to determine the effect of gastrodine on Ox-LDL-stimulated adhesion of HL-60 cells to BAECs. Flow cytometry was used to evaluate the expression of ICAM-1 and VCAM-1 on BAECs. Cytochrome C reduction assay was used to investigate the O2^- level in the cells, and lueiferase reporter gene assay was used to detect the NF-KB transcription activity. Results: As compared with the Ox-LDL-stimulated group, gastrodine significantly decreased the adhesion rate of HL-60 cells to BAECs, the expression levels of ICAM-1 and VCAM-1, and the level and the transcription activity of NF-κB. Conclusion: Gastrodine suppresses Ox-LDL-stimulated adhesion of HL-60 cells to BAECs.
