摘  要:自杀基因前药系统是目前肿瘤基因治疗中的研究热点。其中CD/5-FC自杀基因前药系统是目前研究最多、进展最快的自杀基因系统之一。它对肿瘤细胞的杀死机制除了通过直接毒性作用和旁观者效应以外,还包括自杀基因联合使用、免疫细胞和免疫调节因子的参与、自杀基因与放射治疗联合作用等。本文就CD/5-F自杀基因前药系统的分子生物学基础、抗癌机制以及其在临床实验中面临的问题等研究进展进行综述。Suicide gene therapy is an important method of cancer gene therapy. The suicide gene/prodrug system CD/5-FC is one of the expansively studied and the most advanced suicide gene systems. In addition to the direct toxic effects and the bystander effect, its tumor cell-killing mechanisms include combined use of the suicide gene, the participation of immune cells and immune regulatory factors, and the effects of suicide gene combined with radiotherapy, and so on. The research progress in molecular biological basis and anticancer mechanism of CD/ 5-FC, as well as the problems faced in clinical trials, were reviewed in this article.
