摘 要:目的观察芪蛭益肺颗粒对慢性阻塞性肺疾病(COPD)模型大鼠肺组织转化生长因子-β1(TGF-β1)表达的影响,探讨芪蛭益肺颗粒抑制COPD小气道重塑的机制。方法 72只清洁级Wistar大鼠随机分为空白对照组、模型组和中药治疗组。使用香烟烟雾暴露及气道内滴入脂多糖的联合造模方法建立COPD大鼠模型,于实验第21天起,模型组每日灌胃生理盐水1次,中药治疗组每日灌胃芪蛭益肺颗粒1次,空白对照组正常喂养。各组大鼠于实验第30、40、60天分批处死,留取标本用免疫组化检测肺组织TGF-β1蛋白的表达,实时荧光定量PCR法检测肺组织中TGF-β1mRNA表达。结果模型组中TGF-β1蛋白第30、40、60天表达都较空白对照组升高,第30、40天时与空白对照组比较差异有统计学意义(P<0.05);中药治疗组中TGF-β1蛋白表达在第30、40、60天均低于模型组,其中在第30天时差异具有统计学意义(P<0.05)。模型组中TGF-β1mRNA表达在第40、60天时高于空白对照组,差异具有统计学意义(P<0.05);中药治疗组中TGF-β1mRNA表达在第30、40、60天时较模型组均降低,差异均具有统计学意义(P<0.05)。结论芪蛭益肺颗粒可以通过下调COPD模型大鼠肺组织及小气道上皮TGF-β1的表达来干预COPD小气道重塑。Objective To observe the influence of Qizhi Yifei Granules on expression of transforming growth factor β1 (TGF-β1) of lung tissue in rats with chronic obstructive pulmonary disease (COPD), and discuss the mechanism of Qizhi Yifei Granules in inhibiting small airway remodeling of COPD. Methods Wistar rats (n =72) were randomly divided into blank group, model group and treatment group. The rat model of COPD was established by using the method combining smoking fuming and intratracheal LPS instillation. From the 21st day, model group was given intragastrically saline once a day, treatment group, Qizhi Yifei Granules once a day and blank group, normal feeding. M1 rats were killed on the 30th day, 40th day and 60th day in batches. The expression of TGF-β1 protein was detected by using immunohistoehemistry technique, and expression of TGF-β1 mRNA was detected by using real-time fluorescence quantitative polymerase chain reaction (RT-PCR). Resdts The expression of TGF-β1 protein increased in model group compared with blank group on the 30^th day, 40^th day and 60^th day, and the difference was significant (P 〈 0.05) on the 30^th day and 40^th day. The expression of TGF-β1 protein was lower in treatment group than that in model group on the 30^th day, 40^th day and 60^th day, and the difference was significant (P 〈 0.05) on the 30^th day. The expression of TGF-β1 mRNA was higher in model group than that in blank group on the 40^th day and 60^th day (P 〈0.05). The expression of TGF-β1 mRNA was lower in treatment group than that in model group on the 30^th day, 40^th day and 60^th day (P 〈 0. 05 ). Conclusion Qizhi Yifei Granules can intervene the small airway remodeling of COPD through down-regulating the expression of TGF-β1 of lung tissue and small airway epithelium in rats with COPD.
