摘  要:目的从磷脂酰肌醇(PI)通路探讨脑缺血后抑郁症的发病机制及颐脑解郁方对其干预的作用机理。方法复制脑缺血后抑郁状态大鼠模型后,用酶联免疫法测定PI通路上重要调节因子三磷酸肌醇(IP3)的含量;用蛋白印迹分析法测定PI通路上重要调节因子蛋白激酶C(PKC)的含量的变化及中药颐脑解郁方的干预作用。结果模型组大鼠皮层及海马IP3含量明显升高,与正常组相比有统计学意义(P<0.01)。中药组及西药组大鼠皮层及海马IP3含量均较模型组明显下降,有统计学意义(P<0.01)。与西药组相比,中药组皮层IP3含量无明显差别(P>0.05);中药组海马IP3含量有明显差别,有统计学意义(P<0.05)。模型组、假手术组大鼠海马及皮层PKCα含量明显升高,与正常组相比有显著性差异(P<0.05,P<0.01);经药物治疗后,中药组及西药组大鼠海马及皮层PKCα含量明显下降,与模型组相比有显著性差异(P<0.01)。结论脑缺血后抑郁症的发病可能与PI通路有关;颐脑解郁方和盐酸氟西汀对PI通路有干预作用,这可能是其治疗脑缺血后抑郁症的作用靶点。Objective To investigate the pathogenesis of post-cerebroischemia depression through phosphatidylinositol (PI) signal pathway and intervention mechanism of Yinao Jieyu Fang. Methods After establishing the rat model of post-eerebroischemia depression, the content of inositol triphosphate (IP3) was detected by using enzyme-linked immunosorbent assay (ELISA), and changes of protein kinase C (PKC) and intervention effect of Yinao Jieyu Fang were detected by using Western-blot analysis. Results The content of IP3 in cortex and hippocampus increased significantly in model group compared with normal group ( P 〈 0.01 ) , and decreased significantly in Chinese medicinal group and Western medicinal group compared with model group (P 〈0.01 ). There was no significant difference in content of IP3 in cortex between Chinese medicinal group and Western medicinal group (P 〉 0.05 ) , but there was significant difference in hippocampus (P 〈 0. 05 ). The content of PKCα in cortex and hippocampus increased significantly in model group and sham-operation group compared with normal group (P 〈 0.05 ,P 〈 0.01 ). After treatment, the content of PKCα in cortex and hippoeampus decreased significantly (P〈0. 01). in Chinese medicinal group and Western medicinal group compared with model group Conclusion The onset of post-cerebroischemia depression is related to PI signal pathway. Yinao Jieyu Fang and fluoxertine hydrochloride have intervene effect on PI signal pathway, which may be the effect target of Yinao Jieyu Fang in the treatment of post-eerebroischemia depression.
