摘 要:目的探讨红芪多糖(Hedysariradixpolysaccharide,HRP)对SAM系列快速老化小鼠8(SAMP8)行为学的改善作用和对脑组织中枢神经递质的影响。方法以SAM系列自然老化小鼠1(SAMR1)为对照组,将SAMP8采用随机数字表法分为AD模型组、红芪多糖组、安理申组,连续灌胃给药3个月。实验结束次日,应用Morris水迷宫测试各组小鼠学习、记忆能力。采用高效液相.电化学法检测各组小鼠脑组织中去甲肾上腺素(NE)、多巴胺(DA)、5.羟色胺(5-HT)及5.羟吲哚乙酸(5-HIAA)的含量。结果AD模型组小鼠隐蔽平台逃避潜伏期明显延长,第3、4、5天分别为(55-31±9.62)S、(58.67±10.89)S、(58.45±10.53)s,脑组织中NE为(4.97±1.74)mmol/L、DA为(1.63±0.57)mmol/L、5-HT为(4.29±0.94)mmol/L、5-HIAA为(2.91±0.85)mmol/L,含量均较第1天降低(P均〈0.01)。红芪多糖组小鼠隐蔽平台逃避潜伏期缩短,第3、4、5天分别为(44.73±9.83)S、(40.53±8.37)S、(44.75±9.16)S,脑组织中NE为(7.58±1.62)mmol/L、DA为(3.25±0.70)mmol/L、5.HT为(7.04±0.95)mmol/L、5.HIAA为(4.62±0.79)mmol/L,含量均较第1天增加护均〈O.05)。结论红芪多糖可改善快速老化SAMP8小鼠的学习、记忆能力。Objective To investigate the improving effect of Hedysari radix polysaccharide on the behavior and its influence on cerebral central neurotransmitters in senescence accelerated mouse prone 8 (SAMP 8). Methods The senescence accelerated mice 8 were randomly divided into a model group, a Hedysari radix polysaccharide (HRP) group and a aricept group. SAMR1 were served as the control group. After intragastrical treatment for 3 months, the behavior changes and the levels of norepinephrine(NE), dopamine(DA), 5-hydroxytryptamine (5-HT) and 5-hydroxyindoleacetic acid (5-HIAA) were observed respectively by applying Morris water maze and high performance liquid chromatography (HPLC). Results In the SAMP8 model group, the hidden platform test showed that the latency to find the hidden platform was remarkably prolonged, the latency range from third day to fifth day was (55.31 ±9.62) s, (58.67±10.89) s, (58.45 ±10.53) s, respectively. The level ofNE, DA, 5-HT and 5-HIAA was (4.97±1.74) retool/L, (1.63±0.57) retool/L, (4.29±0.94) mmoUL, (2.91±_0.85)mmol/L respectively, significantly reduced (P〈0.01). Compared with model group, the latency of hidden platform test was (44.73 ±9.83) s, (40.53± 8.37) s, (44.75 ±9.16) s respectively, which were remarkably shortened. The expression level ofNE, DA, 5-HT and 5-HIAA in the HRP treatment group was (7.58±1.62) mmol/L, (3.25 ±0.70) mmol/L, (7.04 ±0.95) mmol/L, (4.62±0.79) mmol/L respectively, which obviously increased (P〈 0.05).
