多重PCR快速鉴定表皮葡萄球菌及mecA耐药基因和icaA生物被膜形成基因

摘  要:目的:建立一种新的多重PCR方法能快速准确地从葡萄菌属中鉴定出表皮葡萄球菌及其mecA基因和icaA基因。方法:针对se2313核酸序列、mecA基因和icaA基因序列设计引物,多重PCR扩增检测。结果:65株(100%)表皮葡萄球菌全部能特异性扩增出se2313核酸序列条带,而其他10种葡萄球菌均未扩增出条带,其中35株(53.8%)表皮葡萄球菌mecA基因扩增阳性,29株(44.6%)表皮葡萄球菌icaA基因扩增阳性。结论:se2313核酸序列能作为鉴定表皮葡萄球菌的特异性靶序列目标,新的多重PCR能快速准确地鉴定出表皮葡萄球菌及其潜在的甲氧西林耐药性和生物被膜形成能力。Objective:To establish a novel multiplex PCR method for rapid and accurate identification of Staphylococcus epidermidis from Staphylococcus with their mecA gene and icaA gene.Methods: The specific primers of se2313 nucleotide sequence,mecA gene and icaA gene were designed for detection by multiplex PCR.Results: The amplification showed that all the 65 Staphylococcus epidermidis strains(100%)were positive for se2313 nucleotide sequence,while the other 10 Staphylococcus strains were negative.Among the 65 Staphylococcus epidermidis strains,thirty-five strains(53.8%)were positive of mecA gene and 29 strains(44.6%)were positive of icaA gene.Conclusion:Se2313 nucleotide sequence is the species-specific target for identification of Staphylococcus epidermidis,the novel multiplex PCR is rapid and accurate to identify Staphylococcus epidermidis species and the potential of methicillin-resistance and producing biofilms.
