摘  要:目的:建立同时测定几种药材中牡荆苷和异牡荆苷含量的方法。方法:采用超高效液相色谱(ultra Performance Liquid Chromatography,UPLC)法,以ACQUITY UPLC BEH C18色谱柱(2.1mm×50mm,1.7μm),甲醇-乙腈-0.2%乙酸水为流动相,流速0.3mL·min-1,梯度洗脱,检测波长340nm,柱温35℃,检测时间7min。检测含羞草、小叶榕、布渣叶、蒲葵、软叶刺葵、淡竹叶、木豆叶和白花败酱草等8种中药材中牡荆苷和异牡荆苷的含量。结果:所建立的测定方法符合液相色谱法要求;牡荆苷、异牡荆苷均在5~250ng(r=0.9999)范围内线性关系良好,平均回收率分别为98.40%、98.35%(n=9),RSD分别为0.43%、0.45%。结论:该法快速简便、准确、分离效果好,能同时快速测定这8种药材中牡荆苷和异牡荆苷含量。Objective To establish an reversed phase-high perform ance liquid chromatography(RP-HPLC) method for simultaneous determination of the contents of vitexin and isovitexin in Herbs. Methods The Ultra Performance Liquid Chromatography(UPLC) system consisting of ACQUITYUPLC BEH C18 column (2.1 mm × 50 mm, 1.7μm) and a solution system of methanol-acetonitrile-0.2 % acetic-acid water as the mobile phase with gradient elution was adopted. The flow rate was set at 0.3 mL/min, the detection wavelength was 340 nm and the column temperature was 35℃, each in 7min. Results The linear ranges of vitexin and isovitexin both were 5-250ng ( r = 0.9999), respectively. The average recoveries (n = 9) were 98.40 % (RSD = 0.43 %), 97.35 % (RSD = 0.45 %), respectively. Conclusion The method is quick, simple and accurate with good reproducibility and can be used for determination and quality control of vitexin and isovitexin in Chinese Medicinal Herbs.
