摘 要:目的:应用高效液相色谱法测定核酸中次黄嘌呤的含量以代替核酸的含量测定。方法:色谱柱为Kromasil C18(4.6 mm×250 mm,5μm);流动相为0.001 mol.L-1磷酸二氢钾溶液(pH6.9);流量1.0 mL.min-1;检测波长260 nm。结果:次黄嘌呤的浓度在2.1036~18.9320 mg.L-1的范围内线性关系良好,回归方程Y=69264.9X+6112.3(r=0.9990)。平均回收率为94.0%,RSD为2.5%。结论:本方法简便、灵敏、准确。Objective: To establish a method for determination of hypoxanthine by HPLC. Method: The determination was performed on a Kromasil Clscolumn, and the mobile phase was 0. 001 mol. L-1KH2PO4and the flow rate was 1.0 mL · min-1. The detection wavelength was at 260 nm. Results: The linear range of hypoxanthine was 2. 1036-18. 9320 mg · L-1 (r =0. 9990) . The average re- covery was 94.0% , and the RSD was 2. 5%. Conclusion: The method was simple, reliable and accurate.
