现代月季离体再生植株影响因素研究

摘  要:以14个现代月季(Rosa hybrida)品种为试验材料,研究基因型、外植体、激素、暗培养及AgNO3对植株再生的影响。结果表明,基因型是影响植株再生的最重要因素,14个现代月季品种中10个品种再生出不定芽,其中Ambiance不定芽分化率最高,叶片为21.05%,叶柄为45.59%;TDZ与IBA激素配合对月季再生的影响比6-BA与NAA激素配合明显;适宜Ambiance叶片再的生最佳培养基为MS+IBA 0.1 mg.L-1+TDZ 1.5 mg.L-1;适宜Ambiance叶柄再生的最佳培养基为MS+IBA 0.05 mg.L-1+TDZ 1.5 mg.L-1;暗培养对提高诱导再生影响效果不明显,但暗培养时间太长将降低诱导率。低浓度AgNO3可提高月季叶片和叶柄不定芽的再生,AgNO3浓度为5 mg.L-1时最利于再生,随着浓度的增加,则会严重抑制月季不定芽分化。The effects of genotype, explant, hormone, duration of dark culture and concentration of AgNO3 on in vitro plant regeneration of 14 rose cultivars (Rosa hybrida) were investigated. The results showed that the genotype was the most important factor of plant regeneration, 10 of 14 rose cultivars could regenerate adventitious shoots, in which Ambiance had the highest differentiation induction rate with 21.05% for leaf and 45.59% for petiole. TDZ combined with IBA was suitable for regeneration than 6-BA combined with NAA. The appropriate media for regeneration from leaf explant of Ambiance was MS+IBA 0.1 mg. L-I+TDZ 1.5 mg L-1; the appropriate media for regeneration from petiole explant of Ambiance was MS+IBA 0.05 mg. L-1TDZ 1.5 rag-L-1. Dark culture did not have many effects on re- generation frequency of the explant, but over long dark culture would decrease induction rate. Low concentration of AgNO3 could increase the regeneration rate of adventitious shoots of leaf and petiole, 5 mg. L-1 AgNO3 was beneficial to differentiation, with the increasing of concentration, AgNO3 will seriously inhibit the differentiation of adventitious shoot of rose.
