加味小苦辛汤对博来霉素A5致肺纤维化小鼠肺组织形态及转化生长因子β_1的影响

摘  要:目的探讨加味小苦辛汤对博来霉素A5(BLM A5)所致肺纤维化动小鼠组织形态及转化生长因子-β1(TGF-β1)表达的影响。方法小鼠腹腔注射BLM A5造模。随机分为正常对照组、模型组、西药组及加味小苦辛汤高、中、低剂量组(中药高、中、低剂量组)。各药物组于造模次日起每日连续给予相应药物,正常对照组与模型组给予羧甲基纤维素灌胃,于给药第14、28日分批随机处死小鼠,观察各组病理切片肺组织形态学改变,免疫组化方法观察肺组织TGF-β1阳性细胞积分光密度和平均光密度。结果模型组较正常对照组细胞积分光密度、平均光密度均显著增加(P<0.01);中药高、中、低剂量组均较模型组显著降低(P<0.01);中药高剂量组较西药组第14日积分光密度差异无统计学意义(P>0.05),第28日较西药组降低(P<0.05),而平均光密度2组差异无统计学意义(P>0.05),且2组较正常对照组增加亦不显著(P>0.05)。结论加味小苦辛汤可显著降低肺纤维化小鼠的肺泡炎程度与肺纤维化程度,降低肺组织细胞TGF-β1的表达。Objective To observe the influence of Modified Xiaokuxin Soup on the lung tissue morphology and TGF-β1 of pulmonary fibrosis mice induced by bleomycin A5(BLM A5).Methods Mice model was made by intraperitoneal injection of BLM A5.Mice were randomly divided into 6 groups:control group,model group,prednisone group and high-,medium-,low-dose of Modified Xiaokuxin Soup groups,which were daily administrated with corresponding drugs after the day of modeling.The mice were killed after 14 and 28 days' consecutive administration,and then the lung tissue morphology,the integral optical density and the average optical density of TGF-β1 were observed.Results The integral optical density and mean optical density of model group significantly increased compared with control group(P 0.01),and that of three dose of Modified Xiaokuxin Soup groups decreased significantly compared with model group(P 0.01). The integral optical density of Modified Xiaokuxin Soup high dose group had no significant difference with western medicine group on the 14th day(P 0.05),and lower than western medicine group on the 28th day(P 0.05),while the difference in the average optical density between the two groups was not significant(P 0.05),and compared with the control group,the increase of the two groups was not significant(P 0.05).Conclusion Modified Xiaokuxin Soup has the effect of anti-inflammatory and anti-fibrosis on pulmonary fibrosis mice,and can reduce the expression of TGF-β1 in lung tissue.
